补坎益离丹慢性毒性初步研究

摘要 【目的】研究长期服用补坎益离丹对小鼠的毒性作用,为犬等动物心阳虚证的治疗和研究提供安全用药依据。【方法】将健康昆明小鼠分为空白组、低剂量组、高剂量组。空白组每天灌胃生理盐水0.5mL、低剂量组每天灌胃给予0.1g/mL补坎益离丹0.5mL、高剂量组每天灌胃给予0.5g/mL补坎益离丹0.5mL,连续60d,观察临床表现。试验结束时摘眼球取血检测血常规、血生化指标,随后脱颈椎处死采集肝脏、心脏、脾脏、肺脏、肾脏,制作组织切片,进行病理组织学观察。【结果】各组小鼠用药期间未见异常临床变化。低剂量组、高剂量组小鼠血常规主要指标如红细胞、白细胞、血小板等均未见明显变化。除低剂量组小鼠的血液中尿素出现显著升高外,低剂量组、高剂量组其他血生化指标均无显著变化。低剂量组和高剂量组肝脏、心脏、脾脏、肺脏、肾脏组织细胞结构均无明显变化。【结论】在长期灌胃给予0.1、0.5g/mL补坎益离丹的情况下,小鼠临床状态良好,血常规主要指标、血生化指标及五脏的组织结构均未见明显的变化,提示补坎益离丹安全性良好,可为犬等动物临床用药研究提供参考。 【Objective】To study the long-term toxicity of Bukanyilidan on mice,and to provide a basis for the treatment and research of heart-yangdeficiency syndrome in dogs and other animals.【Methods】The healthy Kunming mice were divided into blank group,low-dose group and high-dose groups.The blank group was given 0.5 mL saline daily,the low-dose group was given 0.5 mL Bukanyilidan(0.1 g/mL)daily and the high-dose group was given 0.5 mL Bukanyilidan(0.5 g/mL)daily for all 60 days.The clinical symptoms were observed.At the end of the experiment,blood was collected by eyeball extraction for blood routine and blood biochemical indication.Mice were then put to death by cervical spondylolisthesis,and the liver,heart,spleen,lungs and kidneys were taken out to make tissue sections.Subsequently,the changes of pathological histology were observed.【Results】No abnormal clinical changes were observed in all groups.There was no significant change in blood routine parameters such as erythrocyte count,white blood cell count and platelet count in low and high dose groups.There was no significant change in other biochemical indexes except urea increased significantly in low dose group.There were no significant changes in the cellular structure of liver,heart,spleen,lung and kidney in low and high dose groups.【Conclusion】In the longterm administration of Bukanyilidan at 0.1 or 0.5 g/mL,the clinical status of mice is well,and there are no significant changes in the main blood routine indexes,blood biochemical indexes and the tissue structure of liver,heart,spleen,lung and kidney,which suggests that Bukanyilidanis safe and can be used for clinical study in dogs and other animals.
