摘要
【目的】探讨消癌解毒方对结直肠癌细胞增殖的影响及作用机制。【方法】采用细胞计数试剂盒8(CCK-8)观察消癌解毒方含药血清对结直肠癌细胞HT-29、HCT-116、LoVo增殖的影响,采用流式细胞仪观察HCT-116细胞周期分布变化,采用蛋白免疫印迹(Western Blot)法检测HCT-116细胞周期相关蛋白CyclinA2、CyclinB1、CDK1、P21、P27及Notch1/Hes1信号途径相关蛋白Notch1、Hes1的表达。【结果】消癌解毒方含药血清可显著抑制结肠癌细胞HCT-116的增殖,明显改变HCT-116细胞形态,并将细胞周期阻滞于G2/M期,影响周期相关蛋白的表达,抑制Notch1/Hes1信号通路。【结论】消癌解毒方具有抑制结肠癌细胞增殖的作用,其机制可能与阻滞结直肠癌细胞周期,抑制Notch1/Hes1信号通路有关。
Objective To explore the effect of Cancer-Resolving and Toxins-Removing Recipe(CTR)on the proliferation of colorectal cancer cells HT-29,HCT-116,LoVo and its possible mechanism. Methods The effect of CTR-containing serum on the proliferation of colorectal cancer cells HT-29,HCT-116,LoVo was observed by cell count kit 8(CCK-8). The cycle distribution of HCT-116 cells was observed by flow cytometry. The expression levels of cell cycle-associated proteins CyclinA2, CyclinB1, CDK1, P21 and P27, and Notch1, Hes1 in Notch1/Hes1 signaling pathway in HCT-116 cells were detected by Western blotting method. Results The CTRcontaining serum significantly inhibited the proliferation of colorectal cancer HCT-116 cells, altered morphological features of HCT-116 cells,arrested cell cycle in G2/M,interfered the expression of cell cycleassociated proteins,and inhibited the Notch1/Hes1 signaling pathway. Conclusion CTR inhibits the proliferation of colon cancer cells,and its mechanism is possibly related with arresting cell cycle and blocking Notch1/Hes1 signaling pathway.
