摘要 目的:建立高效液相色谱法测定不同产地香加皮药材中杠柳毒苷和4-甲氧基水杨醛的含量。方法:采用Thermo ODS-2 Hypersil色谱柱(250 mmx4.6 mm,5μm),以乙腈-0.05%乙酸(24:76)为流动相;流速1.0 m/min;检测波长220 nm;柱温25 C。结果:杠柳毒苷在0.0068~0.1692 mg/ml(r=0.9999)范围内线性关系良好;4-甲氧基水杨醛在0.0232-0.5807 mg/ml(r=0.9998)范围内线性关系良好。杠柳毒昔平均加样回收率为97.72%,RSD为2.89%(n=9);4-甲氧基水杨醛平均加样回收率为96.63%,RSD为3.61%(n=9)。结论:所建立的高效液相色谱分析方法操作简便,灵敏度高,为评价香加皮质量控制提供可靠依据,为安全用药提供保障。 Objective:To establish a high-performance liquid chromatography(HPLC)method for simultaneous determination of periplocoside and 4一methoxysalicylaldehyde in Periplocae Cortex from different habitats.Aim to establish an effective quality control method for Periplocae Cortex.Methods:The separation was performed on a Themio ODS-2 Hypersil column(250 mm x 4.6 mm,5 jxm)with the isocratic elution of acetonitrile(A)and 0.05%acetic acid(B)(24:76)at the flow rate of 1.0 ml/min.Detection wavelength was set at 220 nm and the column temperature was maintained at 25℃ for the detennination.Results:The results showed that the linear ranges of periplocoside and 4-methoxysalicylaldehyde were 0.0068~0.1692 mg/ml(r=0.9999)and 0.0232~0.5807 mg/ml(r=0.9998),respectively.The average recovery rates for above components were 97.72%(RSD was 2.89%,n=9)and 96.63%(RSD was 3.61%,n=9),respectively.Conclusion:The established HPLC method is simple and accurate for the simultaneous determination of periplocoside and 4-methoxysalicylaldehyde in Periplocae Cortex,and can provide a reliable basis for quality control of Periplocae Cortex and guarantee for safe drug use.
