摘要 目的研究低叶酸环境中赖氨酸去甲基化酶(KDM6A)对神经管畸形(NTDs)产生的影响。方法采用小鼠胚胎干细胞(SV129)、NTD小鼠模型以及NTD人标本;通过甲氨蝶呤(MTX)0.02μmol/L处理SV129细胞24 h,并使用Access 2 Immunoassay系统、竞争性受体结合免疫法测量其叶酸水平;Western blot和免疫荧光检测DNA的断裂及KDM6A表达情况;用小鼠NTD模型,测量其DNA断裂相关修复基因的转录水平;检测人NTD标本中的叶酸含量及KDM6A的表达。结果细胞在低叶酸环境中DNA断裂增加并且KDM6A蛋白表达减少,同时小鼠NTD模型中DNA断裂修复基因KU80/70转录水平表达降低(P<0.05);低叶酸NTD标本的KDM6A与KU80表达也减少。结论低叶酸环境中,DNA断裂增加,KDM6A表达减少。可能是由于启动DNA断裂修复途径异常,导致NTD的发生。 Objective To investigate the effects of lysine-specific demethylase 6A(KDM6A)on neural tube defects(NTDs)in low folate environment.Methods Mouse embryonic stem cells(SV129),NTD mouse models and NTD human specimens were used.SV129 cells were incubated with 0.02μmol/L methotrexate(MTX)for 24 h,and the folate level was measured by Access 2 Immunoassay system and competitive receptor binding immunoassay.DNA fragmentation and KDM6A expression were detected by Western blot and immunofluorescence.The transcription level of the DNA fragment-related repair gene was measured using the mouse NTD model.The folate content and the expression of KDM6A in human NTD specimens were examined.Results In the low folate environment,the DNA fragmentation increased and the expression of KDM6A protein decreased.At the same time,the expression of DNA fragment repair gene KU80/70 in mouse NTD model decreased(P<0.05);the expression of low folate NTD specimen KDM6A and KU80 also decreased.Conclusions In a low folate environment,DNA fragmentation increases and KDM6A expression decreased,which may initiate abnormal DNA repair pathways and lead to NTD.
