冰水环境应激后及恢复期犬生理常数、血清急性期反应物及氧化还原指标的变化 被引量:2

Changes of Physiological Constants,Serum Acute Phase Response Biomarkers and Redox Indexes of Canines after Ice Water Environment Stress and Subsequent Recovery

摘要 为研究冰水环境应激后及恢复期犬生理常数、血清急性期反应物及氧化还原指标的变化,本试验选取健康中华田园犬8条,分别在冰水环境应激前、冰水环境应激结束时(置于冰水0.5 h后)以及冰水环境应激结束后恢复0.5、1.0、3.0、7.0 h时测定体温、心率、呼吸频率,并于颈静脉采血2 mL,测定血清中急性期应激反应物和氧化还原物质的活性或含量。结果显示:与冰水环境应激前相比,体温、心率及血清热休克蛋白70、皮质醇含量与过氧化氢酶活性在冰水环境应激结束时显著降低(P<0.05),血清尿酸、总羰基、丙二醛含量则显著升高(P<0.05);心率及血清维生素E、总羰基、丙二醛含量在冰水环境应激结束后0.5 h时恢复至冰水环境应激前水平(P≥0.05);体温及血清皮质醇、巯基、尿酸含量和铜蓝蛋白活性在冰水环境应激结束后1.0 h时恢复至冰水环境应激前水平(P≥0.05);血清维生素C含量在冰水环境应激结束后3.0 h时恢复至冰水环境应激前水平(P≥0.05)。与冰水刺激结束时相比,犬恢复7.0 h时,其体温、心率及血清皮质醇、维生素E含量显著升高(P<0.05),而呼吸频率及血清超氧化物歧化酶和过氧化氢酶活性与热休克蛋白70、尿酸、总羰基、丙二醛含量显著降低(P<0.05)。由此得出,冰水环境应激可显著改变犬的生理常数、血清应激标志物含量和氧化还原状态,恢复期部分指标仍异常,提示临床上应积极采取保暖、补充抗氧化剂等措施缓解冰水环境应激的危害。 This study was conducted to investigate the changes of physiological constants,serum acute phase response biomarkers and redox indicators of canines after ice water environment stress and subsequent recovery.Eight healthy Chinese rural dogs were selected.The body temperature,heart rate and respiratory rate were measured and 2 mL blood was taken from the jugular vein before ice water environment stress,at the end of ice water environment stress(after 0.5 h in ice water)and recovery 0.5,1.0,3.0,and 7.0 h after the ice water environment stress,respectively.Serum was separated and the activities or contents of acute phase reaction biomarkers and redox substances in serum were determined.The results showed as follows:compared with before ice water environment stress,the body temperature,heart rate and serum heat shock protein 70(HSP70),cortisol contents and catalase activity were significantly decreased at the end of ice water environment stress(P<0.05),while the serum uric acid,carbonyl and malondialdehyde contents were significantly increased(P<0.05).The heart rate,serum vitamin E,total carbonyl and malondialdehyde contents returned to the level of before ice water environment stress after 0.5 h of ice water environment stress(P≥0.05).The body temperature,serum cortisol,sulfhydryl,uric acid contents and ceruloplasmin activity recovered to the level of before ice water environment stress after 1.0 h of ice water environment stress(P≥0.05),while the serum vitamin C content returned to before ice water environment stress level after 3.0 h of ice water environment stress(P≥0.05).Compared with the end of ice water environment stress,the body temperature,heart rate,serum cortisol and vitamin E contents increased significantly(P<0.05),while the respiratory rate,serum total superoxide dismutase,catalase activities and HSP70,uric acid,total carbonyl,malondialdehyde contents decreased significantly when recovery 7.0 h after the ice water environment stress(P<0.05).In conclusion,ice water environment stress can significantly change the physiological constants,serum stress biomarker contents and redox status of canines.Some indicators are still abnormal during the recovery period,which suggested that the clinical measures should be taken to keep warm and supplement antioxidants for controlling the hazards of ice water environment stress.
