复方钩藤口服液的制备及其对小鼠睡眠的改善 被引量:1

摘要 本研究采用正交试验优化复方钩藤口服液的制备工艺,并评价其改善小鼠睡眠的作用。结果表明,优化后的制备工艺为加12倍水提取药材3次,每次2.5 h,以70%乙醇醇沉24 h后浓缩即得。改善小鼠睡眠实验中,高(16 g/kg·BW)、中(8 g/kg·BW)、低(4 g/kg·BW)三个剂量组对小鼠均无直接睡眠作用;与空白对照组相比,中、高剂量组可显著延长戊巴比妥钠诱导的小鼠睡眠时间(P<0.05),而三个剂量组均能显著协同阈下剂量的戊巴比妥钠提高小鼠的入睡率(P<0.05),以及协同巴比妥钠显著缩短小鼠的睡眠潜伏期(P<0.05)。实验结果表明优化的复方钩藤口服液制备工艺稳定、可行,复方钩藤口服液具有改善小鼠睡眠的功能。 In this study,the preparation process of compound Uncaria hirsute Havil.oral liquid(CUHOL)was optimized by the orthogonal experiment and the effect on the sleeping improvement in mice was evaluated.Results showed that:The optimized preparation process was to add 12 times water to estract the medioinal materials for 3 times,2.5 hours each time,and then concentrated after 70%ethanol precipitation for 24 h.The high(16 g/kg·BW),middle(8 g/kg·BW)and low(4 g/kg·BW)doses of compound Uncaria hirsute Havil.oral liquid had no significantly direct sleep effect on mice.Compared with the blank control group,the compound Uncaria hirsute Havil.oral liquid could prolong pentobarbital induced sleeping time at the the middle dose and high dose(P<0.05),meanwhile,the compound Uncaria hirsute Havil.oral liquid not only significantly increased the number of mice that fell asleep in subthreshold pentobarbital sodium induced sleeping time test(P<0.05),but also reduced sleep latency at all dosese(P<0.05).These results showed that the optimized preparation process of compound Uncaria hirsute Havil.oral liquid was stable and feasible,and the compound Uncaria hirsute Havil.oral liquid had the function of improving sleep as well.
