miR-660-3p下调REG4对胃癌细胞增殖、迁移、侵袭的影响及其机制研究 被引量:4

摘要 目的研究miR-660-3p对胃癌MGC-803细胞增殖、迁移和侵袭的影响及潜在的分子机制。方法qRT-PCR检测134例胃癌组织和癌旁组织中miR-660-3p的表达,MTT法和Transwell实验测定过表达miR-660-3p和REG4后MGC-803细胞增殖、迁移和侵袭能力,Western blot检测REG4、Cyclin D1、p21、E-cadherin和MPP-2蛋白的表达,双荧光素酶报告系统验证miR-660-3p与REG4的调控关系。结果与正常癌旁组织相比,胃癌组织中的miR-660-3p表达量显著降低(P<0.05);抑制miR-660-3p可抑制MGC-803细胞增殖、迁移和侵袭,抑制MGC-803中REG4、Cyclin D1和MPP-2表达(P<0.05),促进p21和E-cadherin表达(P<0.05);miR-660-3p靶向负调控REG4的表达;过表达REG4可逆转过表达miR-660-3p对MGC-803细胞增殖、迁移和侵袭的抑制作用。结论miR-660-3p通过靶向REG4抑制MGC-803细胞的增殖、迁移和侵袭。miR-660-3p是胃癌的潜在分子靶点。 Objective To investigate the effect of miR-660-3p on the proliferation,migration and invasion of gastric cancer MGC-803 cells and its potential molecular mechanism.Methods The expression levels of miR-660-3p in 134 gastric cancer tissues and adjacent tissues were detected by qRT-PCR.The proliferation,migration and invasion abilities of MGC-803 cells after overexpression of miR-660-3p and REG4 were measured by MTT and Transwell assay.The expression levels of REG4,Cyclin D1,p21,E-cadherin and MPP-2 proteins in MGC-803 cells were determined by Western blot.The double luciferase reporting system was applied to verify the relationship between miR-660-3p and REG4.Results Compared with adjacent tissues,the expression levels of miR-660-3p in gastric cancer tissues were significantly decreased(P<0.05).Inhibition of miR-660-3p could inhibite the proliferation,migration and invasion of MGC-803 cells,inhibite the expression of REG4,Cyclin D1 and MPP-2(P<0.05),and promote the expression of p21 and E-cadherin in MGC-803(P<0.05).MiR-660-3p targeted targeted negative regulation the expression of REG4.Over-expression of REG4 reversed the inhibitory effect of miR-660-3p over-expression on the proliferation,migration and invasion of MGC-803 cells.Conclusion miR-660-3p inhibits the proliferation,migration and invasion of gastric cancer MGC-803 cells by targeting REG4.miR-660-3p is a potential molecular target for the gastric cancer.
