南伊沟纤维素降解真菌的分离筛选及活性检测 被引量:2

摘要 为从南伊沟常年腐烂树林的腐殖质土壤中分离筛选出高效降解纤维素的真菌,并研究其纤维素降解能力。本实验采用涂布平板法、刚果红纤维素透明圈法、滤纸失重法和滤纸酶活(FPA酶)测定法对其纤维素降解能力进行了初步的测定。筛选得到4株对纤维素具有不同降解活性的菌株,NY10-2,NY16-3,NY17-2和NY19-1,其中菌株NY10-2和NY17-2的降解活性最强,其滤纸失重率分别为49.24%和49.64%,FPA酶活力分别为12.3617IU/mL和11.6348IU/mL。本研究丰富了西藏纤维素降解真菌资源,同时为西藏天然纤维素的处理提供了理论依据和技术支持。 In order to isolate and screen the fungi that degrading cellulose efficiently from the humus soil of the perennial decaying forest in Nanyigou, the cellulose degradation ability was studied in this experiment. The cellulose degradation ability was determined by coating plate method, Congo red cellulose transparent ring method, filter paper weight loss method and filter paper enzyme activity(FPA enzyme) method. Four strains with different degradation activities for cellulose,NY10-2, NY16-3, NY17-2 and NY19-1 were screened. Among them, strains NY10-2 and NY17-2 had the strongest degradation activity, and the weight loss rate of filter paper was 49.24% and 49.64%, respectively, and the FPA enzyme activities were 12.3717 IU/mL and 11.6348 IU/mL, respectively. This paper enriched the cellulose-degrading fungal resources in Tibet and provided theoretical basis and technical support for the treatment of natural cellulose in Tibet.
