青牛解毒汤水煎液对小鼠的镇痛抗炎作用观察 被引量:1

摘要 目的观察青牛解毒汤水煎液对小鼠的镇痛抗炎作用,并探讨其可能的机制。方法将50只小鼠随机分成5组各10只,青牛解毒汤低、中、高剂量组分别以34.125、68.250、136.500 g/kg的青牛解毒汤灌胃,模型组以等体积蒸馏水灌胃,阳性药组以等体积拜阿司匹灵(镇痛)或醋酸地塞米松(抗炎)灌胃,1次/d,连续7 d。采用醋酸扭体实验观察镇痛作用,同时利用二甲苯致小鼠耳廓肿胀、棉球致小鼠肉芽肿实验观察抗急慢性炎症作用。将60只小鼠随机分为6组各10只,青牛解毒汤低、中、高剂量组及阳性药组用药干预同抗炎研究,模型组、空白组等体积蒸馏水灌胃;采用角叉菜胶实验观察小鼠足趾炎性肿胀情况,ELISA法检测足趾肿胀组织中的前列腺素E 2(PGE 2)。结果与模型组比较,青牛解毒汤各剂量组和阳性药组小鼠扭体次数减少、耳肿胀度减轻、棉球肉芽肿形成减少(P<0.05或<0.01)。青牛解毒汤各剂量组和阳性药组足趾肿胀度、组织中PGE 2表达量均低于模型组(P<0.05或<0.01),足趾肿胀度高于空白组(P均<0.01);青牛解毒汤低剂量组和模型组足趾肿胀组织中PGE 2表达量高于空白组(P均<0.01),而青牛解毒汤高剂量组低于空白组(P<0.05)。结论青牛解毒汤具有一定的镇痛抗炎作用,其减轻角叉菜胶所致炎性肿胀作用与降低PGE 2表达有关。 Objective To observe the analgesic and anti-inflammatory effects of Qingniu Jiedu decoction on mice,and to explore its possible mechanism.Methods Fifty mice were randomly divided into five groups.The mice in the low-dose,medium-dose and high-doses Qingniu Jiedu decoction groups were administered 34.125,68.250 and 136.500 g/kg of Qingniu Jiedu decoction.The mice in the model group were treated with equal volume of distilled water,and the positive drug group with an equal volume of aspirin(analgesia)or dexamethasone acetate(anti-inflammatory),once a day,for 7 days.The analgesic effect was observed by acetic acid writhing test,and the anti-acute and chronic inflammatory effects were observed by xylene-induced mouse ear swelling and cotton ball-induced granuloma in mice.Sixty mice were randomly divided into 6 groups.The low-dose,medium-dose and high-dose Qingniu Jiedu decoction groups and the positive drug group were treated with the same method of anti-inflammatory research.The mice of the model group and blank group were administered equal volume of distilled water.The carrageenan experiment was used to observe the inflammatory swelling of the toes of the mice,and the prostaglandin E 2(PGE 2)in the swollen tissues of the toes was detected by ELISA.Results Compared with the model group,the number of writhing decreased,the ear swelling decreased,and the formation of cotton granuloma decreased in Qingniu Jiedu decoction groups and positive drug group(P<0.05 or P<0.01).The swelling degree of the toe and the expression of PGE 2 in the tissues of the Qingniu Jiedu decoction groups and the positive drug group were lower than those in the model group(P<0.05 or P<0.01),and the toe swelling degree was higher than that in the blank group(P<0.01).The expression of PGE 2 in the low-dose Qingniu Jiedu decoction group and the model group was higher than that in the blank group(P<0.01),while the expression of PGE 2 in the high-dose Qingniu Jiedu decoction group was lower than that of the blank group(P<0.05).Conclusion Qingniu Jiedu decoction has certain analgesic and anti-inflammatory effects,which can reduce the inflammatory swelling induced by carrageenan and decrease the expression of PGE 2.
