杜仲醇提取物通过NF-κB信号通路改善大鼠牙周组织病的作用研究 被引量:4

Effect of Eucommiae Cortex Alcohol Extract on Periodontal Disease of Orthodontic Tooth in Rats by NF-κB Signaling Pathway

摘要 目的探讨杜仲醇提取物通过NF-κB信号通路作用于牙周病破骨细胞活化因子(IL-1β)改善大鼠牙周组织病的疗效。方法选取SPF级Wistar大鼠60只,♂,随机数字表法分成4组,正常组,模型组,杜仲醇提取物低、高剂量组,各15只。除正常组外,其他大鼠制备牙周病模型,造模1周后杜仲醇提取物低、高剂量组分别在大鼠上颌右侧第一、第二磨牙间腭、颊侧的龈沟底内注入0.2m L0.1,1.0μg·mg·mL^-1的杜仲醇提取物,正常组与模型组大鼠注入等剂量生理盐水,连续注入5 d。Western-blot、荧光定量PCR检测大鼠牙周膜成纤维细胞内破骨细胞活化因子(IL-1β)蛋白、mRNA表达状况。结果与正常组相比,模型组大鼠成纤维细胞内IL-1β、NF-κB mRNA相对表达量显著上升(P<0.05);与模型组相比,杜仲醇提取物低、高剂量组大鼠成纤维细胞内IL-1β、NF-κB mRNA相对表达量显著下降(P<0.05)。与正常组相比,模型组大鼠血清IL-6、IL-1β及TNF-α含量显著上升(P<0.05);与模型组相比,杜仲醇提取物低、高剂量组大鼠血清IL-6、IL-1β及TNF-α含量显著下降(P<0.05)。与正常组相比,模型组大鼠上颌组织内NF-κB、IL-1β蛋白表达显著上升(P<0.05);与模型组相比,杜仲醇提取物低、高剂量组大鼠上颌组织内NF-κB、IL-1β蛋白表达下降(P<0.05)。结论杜仲醇提取通过NF-κB信号路径对牙周病破骨细胞活化因子(IL-1β)产生作用,降低牙周组织内炎症因子含量,对牙周病起到治疗作用。 OBJECTIVE To analyze the effect of Eucommiae Cortex alcohol extract on periodontal disease osteoclast activating factor(IL-1β)through NF-?B signaling pathway to improve the periodontal disease of orthodontic tooth in rats.METHODS Sixty Wistar male rats of SPF grade were randomly divided into four groups:the normal group,the model group,the low and high doses of Eucommiae Cortex group,15 rats in each group.Except the normal group,other rat model of periodontal disease was prepared.After one week of modeling,the low and high dose groups of Eucommiae Cortex were injected into the bottom of the first and second molars of the right maxillary and the buccal side of the rat with dose of 0.2 mL 0.1,1.0μg·mL^-1 Eucommiae Cortex.Normal group and model group were injected with equal dose of normal saline for 5 d.The expression of osteoclast activating factor(IL-1β)protein and m RNA in rat periodontal ligament fibroblasts was detected by Western-blot and real-time PCR.RESULTS Compared with the normal group,the relative expression of IL-1βand NF-?B m RNA in the fibroblasts of the model group significantly increased(P<0.05).Compared with the model group,the relative expression of IL-1βand NF-?B m RNA in the fibroblasts of low and the high dose of Eucommiae Cortex group decreased(P<0.05).Compared with the normal group,the serum IL-6 and IL-1βin the model group were significantly increased(P<0.05).Compared with the model group,the serum levels of IL-6,IL-1βand TNF-αin the low and the high dose group of Eucommiae Cortex(P<0.05).Compared with the normal group,the expression of NF-?B and IL-1βprotein in the maxillary tissue of the model group significantly increased(P<0.05);compared with the model group,the expression of NF-?B and IL-1βprotein in the maxillary tissue of the rats in the high and low dose of Eucommiae Cortex group was decreased(P<0.05).CONCLUSION Eucommiae Cortex alcohol extraction has a role in the periodontal disease osteoclast activating factor(IL-1β)through NF-?B signal pathway,which reduces the content of inflammatory factors in periodontal tissues and has a therapeutic effect on periodontal disease.
