摘要
目的探讨脂多糖(LPS)对人乳腺癌MDA-MB-231细胞增殖、迁移及侵袭的影响。方法将人乳腺癌MDA-MB-231细胞分为空白对照组和不同浓度(5、10、20、40μg/mL)LPS组,分别培养24、48、72 h取样本,CCK8法检测各组MDA-MB-231细胞增殖率,划痕实验检测各组MDA-MB-231细胞迁移率,Transwell小室实验检测各组MDA-MB-231细胞侵袭能力。结果与空白对照组比较,LPS能促进MDA-MB-231细胞的增殖和迁移(P<0.05),且具有浓度和时间的依赖性,48 h、20μg/mL为最佳时间及浓度,细胞增殖率和迁移率分别为(83.19±3.22)%、(91.59±3.22)%,与空白对照组及LPS 5、10μg/mL组比较差异有统计学意义(P<0.05);空白对照组及LPS 5、10、20、40μg/mL组的细胞侵袭分别为(95.64±4.21)、(119.44±5.44)、(138.99±8.64)、(159.41±9.21)、(143.80±11.45)个,与空白对照组比较,LPS能提高MDA-MB-231细胞的侵袭,以20μg/mL组细胞侵袭个数最多(P<0.05)。结论在体外LPS可促进MDA-MB-231细胞增殖、迁移和侵袭,最佳浓度为20μg/mL。
Objective To observe the effects of lipopolysaccharide(LPS)on the proliferation,migration and invasion of breast cancer MDA-MB-231 cells.Methods The human breast cancer MDA-MB-231 cells were divided into the blank control group and different concentrations LPS groups(0,5,10,20,40μg/mL)and cultured for 24,48,72 h respectively.Then the samples were taken.The proliferation rate of MDA-MB-231 cells in each group was detected by using the CCK8 method.The migration rate and invasion ability of MDA-MB-231 cells in each group were detected by using the scratch test and Transwell chamber assay.Results Compared with the blank control group,LPS could promote the proliferation and migration of MDA-MB-231 cells in a concentration-and time-dependent manner.The optimal time and concentration were 48 h and 20μg/mL.The cell proliferation rate and migration rate were(83.19±3.22)%and(91.59±3.22)%,and the difference was statistically significant compared with the blank control group and the 5,10μg/mL LPS groups(P<0.05).The cell invasion abilities in the blank control group,and 5,10,20,40μg/mL LPS groups were(95.64±4.21),(119.44±5.44),(138.99±8.64),(159.41±9.21),(143.80±11.45)cells respectively,compared with the blank control group,LPS could elevate the invasion ability of MDA-MB-231 cells,the cell invasion numbers was maximal in the 20μg/mL group(P<0.05).Conclusion LPS can promote the proliferation,migration and invasion of breast cancer MDA-MB-231 cells in vitro,and the optimal concentration is 20μg/mL.
