摘要 目的研究MXD3基因在肾透明细胞癌(clear cell renal cell carcinoma,ccRCC)中的表达情况,初步评价其与临床病理学参数之间的关系及对患者预后的影响。方法从癌症基因组图谱数据库(The Cancer Genome Atlas,TCGA)和基因表达汇编数据库(Gene Expression Omnibus,GEO)下载ccRCC中MXD3表达数据和临床病理参数,分析MXD3表达与肾透明细胞癌临床病理学参数之间的相关性及对预后的影响。结果在TCGA数据库(P=0.000)与GEO数据库(P=0.000)中,MXD3在ccRCC组织中的表达量较癌旁组织显著增加,其差异表达可以作为ccRCC的潜在诊断标志物。同时,MXD3表达与T分期(P=0.001)、M分期(P=0.001)、TNM分期(P=0.000)以及病理分级(P=0.001)均密切相关。除此之外,MXD3高表达患者总生存率(overall survival,OS)较低表达患者明显缩短(P=0.000)。单因素(HR=2.287,95%CI:1.670~3.132,P=0.000)和多因素(HR=2.249,95%CI:1.387~3.648,P=0.000)COX回归分析表明MXD3表达是ccRCC患者的独立预后指标。结论在肾透明细胞癌中,MXD3高表达与肿瘤的恶性程度呈正相关,是一个预后不良因素,有望作为判断患者预后的潜在生物学标志物。 Objective To investigate the expression of MXD3 gene in renal clear cell carcinoma(ccRCC)and evaluate its association with clinicopathological parameters and its influence on prognosis.Methods MXD3 gene expression data and clinical information were downloaded from The Cancer Genome Atlas(TCGA)database and Gene Expression Omnibus(GEO)to analyze the association between MXD3 expression and clinicopathological parameters of ccRCC and its influence on prognosis.Results Compared with paracancerous tissues,MXD3 mRNA was upregulated in the tumor tissues in both TCGA(P=0.000)and GEO(P=0.000),and its differential expression was significant in the diagnosis of ccRCC.Meanwhile,MXD3 mRNA expression was associated with several clinicopathological parameters including T stage(P=0.001),M stage(P=0.001),TNM stage(P=0.000),and pathological grade(P=0.001).Kaplan-Meier analysis showed that the overall survival(OS)of patients with high expression of MXD3 was significantly lower than that of patients with low expression(P=0.000).In addition,both univariate(HR=2.287,95%CI:1.670-3.132,P=0.000)and multivariate(HR=2.249,95%CI:1.387-3.648,P=0.000)COX regression models suggested that high expression of MXD3 mRNA was an independent risk factor for the prognosis of patients with ccRCC.Conclusion High expression of MXD3 is negatively correlated with the malignancy of the tumor and is also a poor prognostic factor in patients with ccRCC,which could be a potential biomarker for predicting the prognosis of ccRCC patients.
