儿宝颗粒HPLC指纹图谱研究 被引量:4

摘要 目的建立儿宝颗粒的HPLC指纹图谱。方法采用Agilent ZORBAX Eclipse XDB-C18(4.6 mm×250 mm,5μm)色谱柱;以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱;流速:1.0 mL·min^-1;柱温:30℃;进样量:20μL;检测波长:230 nm。以指纹图谱相似度评价软件结合主成分分析(PCA)、正交偏最小二乘法判别分析(OPLS-DA)评价儿宝颗粒的指纹图谱。结果 HPLC指纹图谱共确定14个共有峰,指纹图谱相似度均在0.900以上,并指认出3个共有成分(芍药苷、柚皮苷、橙皮苷)。模式识别中,11批样品主要按生产年份分为3组,橙皮苷对分组贡献最大。结论所建的方法简便、快速、重复性好,可为儿宝颗粒的整体质量评价提供基础数据。 Objective To establish an HPLC chromatographic fingerprint of Erbao granules.Methods The separation was performed on Agilent ZORBAX Eclipse XDB-C18 column(4.6 mmx250 mm,5 μm) with the mobile phase consisting of acetonitrile and 0.1% phosphoric acid at a flow rate of 1.0 mL·min^-1.The column temperature was 30℃,the detection wavelength was at 230 nm,and the sample injection volume was 20μL.Similarity evaluation software combined with principal component analysis(PCA) and discriminant analysis of orthogonal partial least squares(OPLS-DA) were used to evaluate the HPLC chromatographic fingerprint of 11 batches of Erbao granules.Results There were 14 common peaks in the HPLC chromatographic fingerprint of 11 batches of Erbao granules,and 3 common component(peioside,naringin,hesperidin) were identified.The similar degrees of 11 batches of samples were over 0.900.In pattern recognition,11 batches of samples were mainly divided into 3 groups according to the production year;hesperidin was the component that contributed the most to the grouping.Conclusion This method is simple,fast,and repeatable,and it provides basic data for the overall quality evaluation of Erbao granules.
