摘要
菊粉作为新兴的食品原料被广泛的应用于生物质能源。以菊粉为原料,通过菊粉外切酶处理得到高果糖浆,通过检测还原糖含量确定反应得率,采用DNS法测定还原糖含量。在单因素试验中,以还原糖得率为指标,分别研究了温度、pH、加酶量、底物浓度对还原糖得率的影响。通过单因素试验与正交试验数据分析探究,最优的工艺条件为:酶解温度:45℃,酶解pH=5,底物浓度:50g/L,加酶量:80U/g。在此优化条件下反应60h的还原糖得率为83.63%。本文通过菊粉外切酶将菊粉水解为果糖和葡萄糖,其中果糖为主要产物,葡萄糖为次要产物。重复性试验表明该方案具有良好的复现性,具有重复性好,得率高等优点。可为工业中通过菊粉生产高果糖浆提供一定的指导与参考。
Inulin is widely used as a raw material for biomass in biomass energy.In this paper,the inulin was used as raw material to obtain high fructose syrup by inulin exo-enzyme.The reaction yield was determined by detecting the reducing sugar content,and the reducing sugar content was determined by DNS method.In the single factor experiment,the effects of temperature,pH,enzyme amount and substrate concentration on the yield of reducing sugar were studied.Through the single factor test and orthogonal test data analysis,the optimal process conditions were obtained as follows:enzymatic hydrolysis temperature:45℃,enzymatic hydrolysis pH=5,substrate concentration:50 g/L,enzyme dosage:80 U/g.Under these optimized conditions,the yield of reducing sugars reacted for 60 h was 83.63%.Inulin is hydrolyzed into fructose and glucose by inulin exo-enzyme,in which fructose is the main product and glucose is a secondary product.Repeating tests showed that the scheme has good reproducibility,good repeatability and high yield.It can provide some guidance and reference for the production of high fructose syrup by inulin in industry.
