miR-374b-5p靶向调控ING1表达及对乳腺癌细胞增殖、侵袭和转移的影响 被引量:7

Effects of miR-374b-5p Targeting Regulation ING1 Expression on Proliferation, Invasion and Metastasis of Breast Cancer Cells

摘要 目的研究miR-374b-5p在乳腺癌组织中的表达,探讨miR-374b-5p对乳腺癌细胞增殖、侵袭、转移的影响和对生长抑制因子1(ING1)的靶向调控作用。方法检测miR-374b-5p在50例乳腺癌组织和癌旁组织及乳腺癌细胞系MCF-7、MDA-MB-231、HCC1937和正常乳腺细胞系MCF 10A中的表达水平。选取miR-374b-5p表达水平最高的乳腺癌细胞和正常乳腺细胞,转染miR-374b-5p抑制剂(miR-374b-5p inhibitor),转染48 h后检测2组的miR-374b-5p表达水平,检测细胞增殖率、细胞侵袭和转移能力。研究miR-374b-5p对ING1基因的靶向作用;miR-374b-5p模拟物(miR-374b-5p mimic)对ING1 mRNA表达影响。结果乳腺癌组织中miR-374b-5p表达高于癌旁组织(P<0.05)。miR-374b-5p在乳腺癌细胞系MCF-7、MDA-MB-231、HCC1937的表达高于其在MCF 10A细胞中的表达水平(P<0.05);选取表达水平最高MCF-7细胞用于后续实验。转染48 h,miR-374b-5p inhibitor组miR-374b-5p水平低于对照组(P<0.05)。miR-374b-5p Inhibitor组的细胞增殖率、穿膜细胞数低于对照组(P<0.05);ING1为miR-374b-5p的靶基因,miR-374b-5p mimic组ING1 mRNA的表达减少(P<0.05)。结论miR-374b-5p在乳腺癌组织和细胞系中均高表达,miR-374b-5p可能通过负调控ING1抑制乳腺癌的侵袭和转移。 Objective To study expression of miR-374 b-5 p in breast cancer tissues and to investigate effect of miR-374 b-5 p on proliferation,invasion and metastasis of breast cancer cells and its targeting regulation on growth inhibitory factor1(ING1).Methods Expressions of miR-374 b-5 p in 50 cases of breast cancer tissues and adjacent tissues and breast cancer cell lines MCF-7,MDA-MB-231,HCC1937 and normal breast cell line MCF 10 A were detected.The breast cancer cells with the highest level of miR-374 b-5 p and normal cells of mammary glands were used to transfect.After transfection for 48 h,expressions of miR-374 b-5 p were detected,and the cell proliferation rate,cell abilities of invasion and metastasis were detected in two groups.Targeting effect of miR-374 b-5 p on ING1 gene was studied,and effect of miR-374 b-5 p mimic on ING1 mRNA expression were detected.Results Expressions of mir-374 b-5 p in breast cancer tissues were significantly higher than those in adjacent tissues(P<0.05).Expressions of MCF-7,MDA-MB-231 and HCC 1937 in miR-374 b-5 p breast cancer cell lines were significantly higher than those in MCF-10 A cells(P<0.05),and MCF-7 cells with the highest expression was selected for subsequent experiment.After transfection for 48 h,mir-374 b-5 p expressions in miR-374 b-5 p inhibitor group were significantly lower than those in control group(P<0.05).Proliferation rate and number of membrane penetrating cells in miR-374 b-5 p inhibitor group were significantly lower than those in control group(P<0.05);ING1 was target gene of mir-374 b-5 p,and expression of ING1 mRNA in mir-374 b-5 p mimic group was decreased(P<0.05).Conclusion MiR-374 b-5 p is highly expressed both in breast cancer tissues and cell lines,and miR-374 b-5 p may inhibit invasion and metastasis of breast cancer by negative regulation of ING1.
