醋酸棉酚旋光异构体的制备及其含量测定研究

摘要 目的制备醋酸棉酚旋光异构体并测定其含量。方法采用丙酮-苯胺法从棉籽中制备苯胺棉酚,经乙瞇溶解后在稀硫酸作用下转化,使棉酚游离并与冰醋酸结合制备醋酸棉酚,使用高效液相色谱(HPLC)法测定含量。将自制的醋酸棉酚与1>(一)苏-1-对硝基苯基-2-氨基-1,3-丙二醇进行衍生化反应,衍生物经硅胶柱色谱分离,分别制得(+)和(一)-醋酸棉酚衍生物,采用盐酸水解法对(+)和(一)-醋酸棉酚衍生物进行水解,分别制备(+)-醋酸棉酚和(一)-醋酸棉酚单体,使用HPLC法测定含量。结果自制醋酸棉酚的含量为90.3%,衍生化反应和硅胶柱色谱可有效制备分离(+)和(一)-醋酸棉酚衍生物,水解后制备的(+)-醋酸棉酚和(一)-醋酸棉酚含量分别为90.8%、91.5%。结论本方法操作简便、重现性好,适用于棉酚旋光异构体的制备及含量测定。 Objectives Preparation and determination of gossypol acetate optical isomer・Methods The aniline gossypol was prepared from cottonseed by acetone aniline method・After dissolving in ether,gossypol was transformed by dilute sulfuric acid・Gossypol was dissociated and combined with glacial acetic acid to prepare gossypol acetate・The content of gossypol acetate was determined by HPLC・The derivative reaction of gossypol acetate and D-(—)su-l-p—nitrophenyl-2-amino-l,3-propanediol was carried out.The derivatives were separated by silica gel column chromatography,and the derivatives of(+)and(—)-gossypol acetate were prepared,respectively.The derivatives of(+)-gossypol acetate and(—)-gossypol acetate were hydrolyzed by hydrochloric acid hydrolysis method,and the content was determined by HPLC.Results The content of gossypol acetate was 90.3%・Derivatization reaction and silica gel column chromatography can effectively separate(+)and(—)-gossypol acetate derivatives・The contents of(—)-gossypol acetate and(—)-gossypol acetate were 90.8%and 91.5%,respectively.Conclusion The method is simple,reproducible and suitable for the preparation and determination of gossypol optical isomers.
