加味大黄牡丹皮汤对急性盆腔炎模型大鼠子宫、卵巢的保护作用及NF-κB信号通路的影响 被引量:6

Protective effect of Jiawei Dahuang Mudan Pi Decoction on uterus and ovary and its effect on NF-κB signal pathway in rats with acute pelvic inflammatory disease

摘要 目的探讨加味大黄牡丹皮汤对急性盆腔炎模型大鼠子宫、卵巢的保护作用及NF-kB信号通路的影响。方法取10只大鼠为对照组,另取30只大鼠建立盆腔炎大鼠模型,将大鼠按随机数字表法随机分为模型组(混合菌液接种法制备)、加味大黄牡丹皮汤组(加味大黄牡丹皮汤6.0 g/kg)、阿奇霉素组(阿奇霉素1.7 mg/kg)o第1次感染8 d后,收集大鼠外周血、子宫、卵巢,酶联免疫吸附试验检测血清中IL-6、IL-8、TNFp的含量;右侧子宫和卵巢HE染色观察组织结构和病变特点,并评估炎症细胞浸润程度;蛋白质印迹法和逆转录聚合酶链反应检测NF-KBp65 JkB«相对蛋白表达水平。结果与对照组比较,模型组大鼠子宫和卵巢病理病变明显,可见大量炎症细胞浸润,炎症细胞的组织学半定量评分升高,血清中IL-6、IL-8、TNF-a含量升高,子宫和卵巢组织中NF-KBp65 JKBa相对蛋白表达水平上调,差异有统计学意义(P V0.05)。与模型组比较,加味大黄牡丹皮汤组和阿奇霉素组大鼠炎症细胞浸润情况相对改善,组织学半定量评分降低,血清中IL-6、IL-8、TNF-a含量降低,子宫和卵巢组织中NF-KBp65 JKBa相对蛋白表达水平降低,差异有统计学意义(P<0.05).结怡加味大黄牡丹皮汤通过减少炎症细胞的浸润和抑制细胞因子IL-6,IL-8,TNF-a的过量产生,对急性盆腔炎大鼠子宫、卵巢有一定的保护作用。 Objective To investigate the protective effect of Dahuang Mudan Decoction on uterus and ovary and its effect on NF-kB signal pathway in rats with acute pelvic inflammatory disease.Methods 10 rats were taken as the blank control group,another 30 rats were taken to establish the model o£pelvic inflammation and then randomly divided into model group(prepared by mixed bacterial inoculation),Dahuang Mudan Decoction group (Dahuang Mudan Decoction (6.0 g/kg) , azithromycin group (azithromycin 1.7mg/kg) according to the random number table. At the eight h day after the first infection, the peripheralblood, uterus and ovary of rats were collected・ The levels of IL-6, IL-8 and TNF-a in serum were detectedby enzyme-linked immunosorbent assay (ELISA)・ The tissue structure and pathological characteristics ofthe right uterus and ovary were observed by HE staining, and the infiltration degree of inflammatory cellswas evaluated. The expression levels of NF kbp65 , IwBa protein and mRNA were detected by Western blotand reverse transcription polymerase chain reaction・ Results Compared with the control group, the pathologicalchanges of uterus and ovary in the model group were obvious;a large number of inflammatory cellsinfiltrated;the histological semi-quantitative scores of inflammatory cells was significantly increased;thecontents of IL-6 , IL-8 and TNF-a in serum significantly increased;and the expression levels of NF-icBp65,IkBo protein and mRNA in uterus and ovary tissue significantly increased・ The difference were statisticallysignificant (P <0.05)・ Compared with the model group, the Inflammatory cell infiltration in Dahuang MudanDecoction group and Az让hromycin group were improved, semi-quantitative histological score was decreased,serum levels of IL-6, IL-8 and TNF-a were significantly decreased, and expressions o£ NF-icBp65,IwBa protein and RNA in uterus and ovary were significantly decreased. The difference were statisticallysignificant (P V0.05). Conclusion Dahuang Mudan Decoction has protective effect on acute pelvic inflammatoryrats by reducing the infiltration of inflammatory cells and inhibiting the excessive production of cytokinesIL-6, IL-8 and TNF-a.
