CSRP2在成人非M3-急性髓系白血病患者骨髓中的表达

Expression of CSRP2 in bone marrow of adult non-M3 acute myeloid leukemia patients

目的:探讨成人非M3-急性髓系白血病(M3-AML)患者CSRP2基因表达的特点及临床意义。方法:应用实时荧光定量PCR法检测20名同期健康造血干细胞移植(HSCT)供者(正常对照)及30例初治成人非M3-AML患者骨髓中CSRP2 mRNA的表达水平。30例中男18例,女12例,中位年龄33岁;经治疗均达完全缓解,均有完整的临床资料及随访资料。结果:非M3-AML患者CSRP2 mRNA表达水平高于正常对照(P=0.009)。将非M3-AML患者以CSRP2 mRNA表达水平中位数为界点分为低、高表达组。两组年龄、性别、初诊时白细胞计数、血小板计数、骨髓原始细胞比例、ETO融合基因表达、FLT3-ITD突变、NPM1突变、预后危险度分层、HSCT患者比例等差异均无统计学意义(P>0.05)。Kaplan-Meier生存分析结果显示,两组无复发生存曲线差异有统计学意义(χ^2=4.384,P=0.036),CSRP2 mRNA高表达组患者预后好于低表达组。结论:CSRP2高表达的初治成人非M3-AML患者预后更好;CSRP2可能成为成人非M3-AML患者的新的预后分子标志物。

Aim:To investigate the expression and clinical significance of CSRP2 gene in adult non-M3 acute myeloid leukemia(non-M3-AML).Methods:The CSRP2 mRNA expression in bone marrow of 20 healthy hematopoietic stem cell transplant(HSCT)donors at the same time(normal controls)and 30 newly diagnosed adult non-M3-AML patients were detected by real-time quantitative PCR.Among the 30 patients,18 were male and 12 were female,with a median age of 33 years.All the 30 patients had complete clinical data and follow-up data,and were completely relieved after treatment.Results:The CSRP2 mRNA expression of non-M3-AML patients was significantly higher than that of the normal controls(P=0.009).The 30 patients were divided into low and high expression groups according to the median expression level of CSRP2 mRNA as the boundary point.There were no significant differences in age,gender,leukocyte count,platelet count,proportion of bone marrow primordial cells,expression of ETO fusion gene,FLT3-ITD mutation,NPM1 mutation,prognosis risk stratification and proportion of HSCT patients between the two groups(P>0.05).The results of Kaplan-Meier survival analysis showed that the difference in relapse free survival curves between the two groups was statistically significant(χ^2=4.384,P=0.036),the prognosis of patients with high CSRP2 mRNA expression was better than those with low expression.Conclusion:Adult non-M3-AML patients with high CSRP2 expression may have better prognosis;CSRP2 may be a new prognostic molecular marker for adult non-M3-AML patients.
