低弹性模量医用钛合金梯度复合材料对小鼠成纤维细胞L929增殖的影响

摘要 目的:探索低弹性模量医用钛合金梯度复合材料对小鼠成纤维L929细胞形态及增殖率的影响。方法:制备低弹性模量医用Ti-27Nb-6Zr-5Mo合金(钛合金)试件、经氧化处理的钛合金(氧化层)试件、经氧化处理并在表面等离子喷涂钽涂层的钛合金(钽涂层)试件。利用3种试件的浸提液体外培养L929细胞,分为钛合金组、氧化层组和钽涂层组,同时以用含体积分数10%胎牛血清的RPMI 1640培养基培养的细胞作为阴性对照组,以体积分数0.6%苯酚溶液培养的细胞作为阳性对照组。细胞培养第1、3、5天,倒置相差显微镜下观察细胞形态,采用MTT法检测细胞的增殖率。结果:钛合金组、氧化层组和钽涂层组L929细胞形态均正常,生长良好;同一时间点各组间A值比较差异有统计学意义(P<0.05),钽涂层组A值最大;钛合金组、氧化层组、钽涂层组和阴性对照组A值均随着培养时间的延长而增加(P<0.05);低弹性模量医用钛合金梯度复合材料试件毒性分级为0或1级。结论:低弹性模量医用钛合金梯度复合材料可促进L929细胞增殖且无细胞毒性。 Aim:To explore the effects of the medical titanium alloy gradient composites with low elastic modulus on the morphology and proliferation of L929 cells.Methods:L929 cells were cultured in vitro with leaching liquids of low elastic modulus medical titanium alloy(titanium alloy group),low elastic modulus medical titanium alloy by oxidation treatment(oxide layer group),and low elastic modulus medical titanium alloy with tantalum coatings by plasma spraying after oxidation treatment(tantalum coating group)for 1,3 and 5 days.At the same time,L929 cells cultured with RPMI 1640 containing volume fraction 10%fetal bovine serum and volume fraction 0.6%phenol were served as the negative and positive control.The cell morphology was observed under inverted phase contrast microscope,and the proliferation was detected by MTT method.Results:L929 cells in titanium alloy,oxide layer and tantalum coating groups showed normal morphology and vigorous growth;at the same time point,there was significant difference in A value between the 3 groups(P<0.05),and the A value of tantalum coating group was the maximum;A value of titanium alloy,oxide layer,tantalum coating and negative control groups increased with the incubation time(P<0.05).The toxicity grade of three test pieces was 0 or 1.Conclusion:The medical titanium alloy gradient composites with low elastic modulus has no cytotoxicity.
