苦瓜总皂苷的响应面法提取条件及降血糖活性的研究 被引量:5

Optimization of extraction of Total saponins of Momordica charantia L.using response surface methodology and its antioxidative activity in vitro

摘要 目的研究苦瓜总皂苷的最佳提取条件及其对a-葡萄糖苷酶体外抑制活性,为苦瓜皂苷降血糖活性的深入研究提供参考。方法建立苦瓜总皂苷的响应面模型,利用AB-8大孔吸附树脂纯化苦瓜皂苷粗提物,并以苦瓜皂苷对a-葡萄糖苷酶的体外抑制活性为考察指标,考察不同剂量苦瓜皂苷的降血糖活性。结果当乙醇浓度73.85%、料液比1:20、提取温度78.59℃、提取时间1.82h时,苦瓜总皂苷的预测质量分数为14.07%,验证质量分数为13.98%。经纯化后苦瓜皂苷的含量由13.98%提高到65.94%,且在一定的浓度范围内其对a-葡萄糖苷酶的抑制作用呈现出良好的剂量-效应关系。结论该实验研究为苦瓜皂苷的后期研究提供了理论依据。 Objective To study the optimal extraction conditions of total saponins of Momordica charantia L.and its inhibitory activity against a-glucosidase in vitro,and provide reference for the in-depth study of the hypoglycemic activity of Momordica saponins.Methods The response surface model of total saponins of Momordica charantia L.was established.The crude extract of Momordica saponin was purified by AB-8 inacroporous adsorption resin.The in vitro inhibitory activity of Momordica saponin on a-glucosidase was used as an indicator to investigate the different closes of Momordica saponin.Hypoglycemic activity.Results When the ethanol concentration was 73.85%,the ratio of material to liquid was 1:20,the extraction temperature was 78.59℃,and the extraction time was 1.82 h,the predicted mass fraction of total saponins of bitter gourd was 14.07%,and the verification mass fraction was 13.98%.After purification,the content of Momordica saponin increased from 13.98%to 65.94%,and its inhibition of a-glucosidase showed a good dose-effect relationship within a certain concentration range.Conclusion This experimental study provides a theoretical basis for the later study of bitter melon saponins.
