贝母素乙抑制结肠癌细胞生长及裸鼠成瘤 被引量:1

Inhibitive Effect of Peiminine on the Growth of Colon Cancer Cells and Tumor Formation in Nude Mice

摘要 目的研究贝母素乙对结肠癌细胞生长以及裸鼠成瘤的影响.方法体外试验检测贝母素乙对结肠癌SW480细胞增殖和侵袭的影响,以及Ki67、PCNA、VEGF、E-cadherin、N-cadherin、HIF-1α、Akt/p-Akt表达水平;构建结肠癌移植瘤裸鼠模型,分析贝母素乙体内抗肿瘤活性及HIF-1α、VEGF、Akt/p-Akt表达影响.结果贝母素乙能够明显抑制SW480细胞克隆增殖和侵袭(P<0.05),降低Ki67、PCNA、N-cadherin、E-cadherin、HIF-1α、p-Akt/Akt表达量(P<0.05),升高Vimentin表达水平(P<0.05),还可以抑制肿瘤增长,抑制移植瘤组织Ki67、VEGF、HIF-1α、p-Akt/Akt表达(P<0.05).结论贝母素乙可能通过抑制Akt信号通路,抑制结肠癌SW480细胞克隆增殖、侵袭和裸鼠移植瘤的生长. Objective To study the effects of Peiminine on the growth of colon cancer cells and tumor formation in nude mice.Methods The effects of Peiminine on the proliferation and inva-sion of colon cancer SW480 cells were detected.The expression levels of Ki67,PCNA,VEGF,E-cadherin,N-cadherin,HIF-1αand Akt/p-Akt were detected by Western blotting.The nude mouse models of colon cancer xenografts were constructed,and the effects of Peiminine on the antitumor activity and the expression levels of HIF-1α,VEGF and Akt/p-Akt were analyzed.Results Peimi-nine could significantly inhibit the cell clone proliferation and invasion of SW480 cells in a signifi-cantly dose-dependent manner(P<0.05),reduce the expression levels of Ki67,PCNA,N-cad-herin,E-cadherin,HIF-1αand p-Akt/Akt(P<0.05),and increase the expression level of Vi-mentin(P<0.05),and inhibit tumor growth and the expression of Ki67,VEGF,HIF-1αand p-Akt/Akt in xenograft tissues.Conclusion Peiminine can inhibit the clone proliferation and invasion of colon cancer SW480 cells,and inhibit the tumor growth of xenograft nude mouse models via the Akt signaling pathway.
