摘要
溶质转运蛋白家族相关基因在黑色素合成过程中发挥着重要作用。为了进一步证明新发现的SLC6A9和SLC38A4两个候选基因在黑色素合成/沉积过程中的作用,以小鼠黑色素瘤B16细胞作为受试细胞,通过siRNA抑制SLC6A9和SLC38A4基因表达,分析相关黑色素合成关键基因表达,进一步确定SLC6A9和SLC38A4是否参与黑色素合成与调控。结果表明:抑制SLC6A9表达能显著下调PMEL、KIT和TYRP1基因表达(P<0.01),抑制SLC38A4基因表达能显著下调MC1R基因的mRNA表达水平(P<0.01),说明SLC6A9和SLC38A4基因表达可能参与B16细胞黑色素合成,其中SLC38A4基因可能通过MC1R信号通路调控真黑色素和褐黑色素合成比例。
The Solute Carrier(SLC)family group of membrane transporter genes has been identified to play an important role in melanogenesis.The experiment was conducted to investigate the role of two candidate genes(SLC6A9 and SLC38A4)in melanogenesis of murine melanoma B16 cell line.SLC6A9 and SLC38A4 genes were knocked down by small interfering RNA(siRNA)fragments,and then,the melanogenesis-related genes expression was detected using qRT-PCR.The results showed that the PMEL,KIT and TYRP1 mRNA level was significantly reduced when the SLC6A9 mRNA was knocked down by siRNA(P<0.01).The MC1R mRNA level was significantly reduced when the SLC38A4 mRNA was knocked down by siRNA(P<0.01).It was concluded that SLC6A9 and SLC38A4 mRNA levels had a positive correlation with melanogenesis of murine melanoma B16 cell line,SLC38A4 may regulate the ratio of eumelanin to pheomelanin by MC1R signaling pathway.
