miRNA-107在胃癌、结直肠癌及其癌前病变中的表达水平及临床意义

Expression and clinical significance of miRNA-107 in gastric cancer, colorectal cancer and its precancerous lesions

[目的]研究miRNA-107在胃癌、结直肠癌及其癌前病变中的表达水平及临床意义。[方法]选择2017年5月~2018年5月北京医院收治的胃癌及结直肠癌患者各60例,另外选择健康受试者、慢性萎缩性胃炎、慢性萎缩性胃炎伴异型增生、炎症性息肉和腺瘤性息肉患者各15例进行研究,收集受试者的病理活检标本及血清标本。采用RT-PCR检测受试者血清和组织中的miRNA-107表达情况。[结果]胃癌患者血清中的miRNA-107水平(14.67±1.78)显著高于其他受试者(P<0.05);慢性萎缩性胃炎伴异型增生患者血清miRNA-107水平(3.65±0.45)显著高于健康受试者(1.03±0.08)和单纯慢性萎缩性胃炎患者(1.43±0.19)(P<0.05)。胃癌组织中的miRNA-107水平(23.89±2.92)显著高于其他组织(P<0.05);慢性萎缩性胃炎伴异型增生组织中miRNA-107水平(6.78±0.83)显著高于单纯慢性萎缩性胃炎组织(2.23±0.27)(P<0.05)。结直肠癌患者血清中的miRNA-107水平(18.98±2.32)显著高于其他受试者(P<0.05);腺瘤性息肉患者血清miRNA-107水平(5.89±0.72)显著高于健康受试者(1.01±0.10)和炎症性息肉患者(2.56±0.31)(P<0.05)。结直肠癌组织中的miRNA-107水平(28.96±3.54)显著高于其他组织(P<0.05);腺瘤性息肉组织miRNA-107水平(8.75±1.08)显著高于炎症性息肉组织(3.47±0.42)(P<0.05)。随着癌症临床分期的升高,miRNA-107在胃癌和结直肠癌患者血清和组织中均呈现升高趋势。[结论]miRNA-107水平在胃癌和结直肠癌中为高表达,并且可能与疾病进展有关,当患者的胃和结直肠具有癌变倾向时,miRNA-107水平会相应升高。

[Objective]To reveal the expression level and clinical significance of miRNA-107 in gastric cancer, colorectal cancer and its precancerous lesions.[Methods]From May 2017 to May 2018,patients with gastric cancer(n=60)and colorectal cancer(n=60)admitted to Beijing Hospital were selected.In addition,15 healthy subjects,15 patients with chronic atrophic gastritis,15 patients with chronic atrophic gastritis with dysplasia,15 patients with inflammatory polyps and 15 patients with adenomatous polyps were selected,and pathological biopsy specimens and serum samples were collected from all subjects.The expression of miRNA-107 in serum and tissues of the subjects was detected by RT-PCR.[Results]The levels of miRNA-107 in serum of patients with gastric cancer(14.67±1.78)were significantly higher than those of other subjects(P<0.05).The level of serum miRNA-107(3.65±0.45)in patients with chronic atrophic gastritis with dysplasia was significantly higher in healthy subjects(1.03±0.08)and patients with chronic atrophic gastritis(1.43±0.19)(P<0.05).The level of miRNA-107 in gastric cancer tissues(23.89±2.92)was significantly higher than that in other tissues(P<0.05).The level of miRNA-107 in chronic atrophic gastritis with dysplasia tissues(6.78±0.83)was significantly higher than that in chronic atrophic gastritis tissue(2.23±0.27)(P<0.05).The level of miRNA-107 in serum of patients with colorectal cancer(18.98±2.32)was significantly higher than that of other subjects(P<0.05);the level of serum miRNA-107 in patients with adenomatous polyps(5.89±0.72)was significantly higher than that of healthy subjects(1.01±0.10)and patients with inflammatory polyps(2.56±0.31)(P<0.05).The level of miRNA-107 in colorectal cancer tissues(28.96±3.54)was significantly higher than that in other tissues(P<0.05);the level of miRNA-107 in adenomatous polyp tissues(8.75±1.08)was significantly higher than that in inflammatory polyp tissue(3.47±0.42)(P<0.05).With the increase of clinical stage of cancer,miRNA-107 showed an increasing trend in serum and tissues of patients with gastric cancer or colorectal cancer.[Conclusion]The level of miRNA-107 is highly expressed in gastric cancer and colorectal cancer and may be associated with disease progression.When the stomach and colorectal of patients have a tendency to become cancerous,the level of miRNA-107 will increase accordingly.
