摘要
目的:探讨表儿茶素抗高糖高脂诱导H9C2细胞死亡的作用。方法:将H9C2细胞分为正常对照(Control)组、高糖高脂(HGHL)组、溶剂对照(DMSO)组、表儿茶素预处理(EC)组,后两组分别用DMSO和表儿茶素预处理2 h之后再用高糖高脂培养基培养24 h。采用CCK-8法检测细胞存活率;倒置显微镜观察细胞形态变化;Hoechst 33258染色观察细胞死亡及核变化情况;化学比色法测定乳酸脱氢酶(LDH)和超氧化物歧化酶(SOD)活性、还原型谷胱甘肽(GSH)含量。结果:与HGHL组比较,EC组细胞存活率明显提高(P<0.05);细胞形态较稳定;还原型GSH含量及SOD活性均明显升高(P<0.05),LDH活性明显降低(P<0.01)。结论:表儿茶素对高糖高脂诱导的H9C2细胞死亡有明显的抑制作用。
Objective:To explore the effect of epicatechin against H9C2 cell death induced by high glucose and high lipid.Methods:H9C2 cells were divided into normal control group,high glucose and high lipid(HGHL)group,DMSO group and epicatechin pretreatment(EC)group.The latter groups were treated with DMSO and epicatechin was pretreated for 2 h and then incubated with high glucose and high lipid for 24 h.Cell viability was detected by CCK-8 method;cell morphology was observed by inverted microscope;cell death and nuclear changes were observed by Hoechst 33258 staining;lactate dehydrogenase(LDH)and superoxide dismutase(SOD)activities,reduced glutathione(GSH)content were determined by chemical colorimetry.Results:Compared with the HGHL group,the cell survival rate of the EC group was significantly increased(P<0.05);the cell morphology was stable;the reduced GSH content and SOD activities were significantly increased(P<0.05),and the LDH activity was significantly decreased(P<O.Ol).Conclusion:Epicatechin has a significant inhibitory effect on H9C2 cell death induced by high glucose and high lipid.
