circCMTM3通过海绵mi R-588调控细胞迁移和侵袭 被引量:2

circCMTM3 Regulates Cell Migration and Invasion Through Sponging miR-588

摘要 目的:探讨环状RNA(circRNA)circCMTM3在骨肉瘤细胞中的表达及其作用机制。方法:利用qRT-PCR检测miRNA和circRNA的表达,CCK-8检测细胞增殖,Transwell实验检测细胞迁移和侵袭,流式细胞术检测细胞周期和凋亡,萤光素酶报告基因检测circCMTM3与miR-588的靶向结合。结果:circCMTM3在骨肉瘤细胞中高表达,敲除circCMTM3显著抑制骨肉瘤细胞的生长、迁移和侵袭,促进细胞凋亡;过表达circCMTM3促进骨肉瘤细胞的迁移和侵袭,miR-588过表达抑制细胞的迁移和侵袭,而过表达circCMTM3通过海绵作用抑制miR-588的功能。结论:circTMCM3在骨肉瘤细胞中高表达,并通过海绵miR-588促进骨肉瘤进展,这项研究可能为骨肉瘤提供新的潜在治疗途径。 Objective:To investigate the expression and function mechanism of circCMTM3 in osteosarcoma cells.Methods:The expressions of miRNA and circRNA were detected by qRT-PCR.CCK-8 was used to detect cell proliferation.Transwell assay was used to detect cell migration and invasion.Cell cycle and apoptosis were detected by flow cytometry.Luciferase reporter gene assay was used to detect whether circCMTM3 bind with miR-588.Results:circCMTM3 was highly expressed in osteosarcoma cells,and circCMTM3 knockout significantly inhibited the growth,migration and invasion of osteosarcoma cells,and promoted apoptosis.Overexpression of circCMTM3 promoted the migration and invasion of osteosarcoma cells,and overexpression of miR-588 inhibited the ability of migration and invasion in osteosarcoma cells,while overexpression of circCMTM3 inhibited the function of miR-588 through sponge action.Conclusion:circTMCM3 is highly expressed in osteosarcoma cells and promotes progression of osteosarcoma through sponge miR-588.This study may provide new potential therapeutic pathways for osteosarcoma.
