摘要
目的:探讨异甘草素联合骨髓间充质干细胞在脑梗死大鼠中的保护作用,为脑梗死治疗提供思路。方法:采用线栓法完成急性脑梗死大鼠模型,随机分为异甘草素组、骨髓间充质干细胞组、联合干预组,比较各组神经功能评分、HE染色结果、脑梗死面积、炎症因子水平。结果:联合干预组干预后NIHSS评分均低于骨髓间充质干细胞组、异甘草素组(P<0.05);骨髓间充质干细胞组干预后NIHSS评分均低于异甘草素组(P<0.05);骨髓间充质干细胞组脑梗死细胞轮廓清晰,胞质丰富,可见明显坏死细胞;联合干预组细胞轮廓清晰,胞质丰富,可见少许坏死细胞;联合干预组干预后脑梗死体积,均低于骨髓间充质干细胞组与异甘草素组(P<0.05);联合干预组干预后4周TNF-α及IL-1β水平低于骨髓间充质干细胞组与异甘草素组(P<0.05);联合干预组干预后4周IL-10水平高于骨髓间充质干细胞组与异甘草素组(P<0.05)。结论:异甘草素联合骨髓间充质干细胞用于脑梗死大鼠中有助于改善神经功能,减少脑梗死面积。
Objective:To explore the mechanism of isoglycyrrhiza combined with bone marrow mesenchymal stem cells in rats with cerebral infarction,and to provide ideas for the treatment of cerebral infarction.Methods:Rat models of acute cerebral infarction were established by thread embolization,and were randomly divided into model control group,bone marrow mesenchymal stem cells(BMSCs)group and combined intervention group.Neurological function scores,HE staining results,cerebral infarction area and levels of inflammatory factors in each group were compared.Results:The NIHSS scores of the combined intervention group were lower than those of the BMSCs group and the isoliquiritigenin group(P<0.05);the NIHSS scores of the BMSCs group after intervention were lower than those of the isoliquiritigenin group(P<0.05);the cerebral infarction cells in the BMSCs group had clear outline and abundant cytoplasm,and necrotic cells could be clearly seen.The volume of cerebral infarction in the combined intervention group was lower than that in the BMSCs group and isoliquiritigenin group(P<0.05).Conclusion:isoglycyrrhiza combined with bone marrow mesenchymal stem cells can improve neurological function and reduce the area of cerebral infarction in rats with cerebral infarction.
