摘要
研究转染沉默IGFBP-2抑制食管癌细胞系TE-1增殖、转移的影响。通过构建好的IGFBP-2的SiRNA绿色荧光载体转染到人食管癌细胞系TE-1中,于荧光显微镜下观测转染效率,通过Western-blot证实基因沉默的效果,并通过CCK8,Transwell和流式来分别评估IGFBP-2-SiRNA对细胞增殖,侵袭和凋亡的影响。结果显示:Western-blot法证实IGFBP-2能有效地被SiRNA沉默,而且IGFBP-2-SiRNA能抑制食管癌细胞系TE-1的增殖\,侵袭和诱导凋亡。实验结果表明IGFBP-2在食管癌的发病机理中可能有着重要的作用。IGFBP-2也许能成为食管癌治疗的一个靶向基因。
This paper is to study the effect of transfecting and silencing gene IGFBP-2 on inhibition of proliferation and metastasis of esophageal carcinoma cell line TE-1.By transfecting IGFBP-2 to esophageal carcinoma cell line TE-1 through the ready SiRNA green fluorescent carrier,the transfection efficiency was observed under a fluorescence microscope,the effect of gene silencing was verified by means of western-blot,and the effect of IGFBP-2-SiRNA on cell proliferation,invasion and apoptosis was evaluated respectively with CCK8,Transwell and flow cytometry.The western-blot method confirmed that IGFBP-2 could be effectively silenced by SiRNA,and that IGFBP-2-SiRNA could inhibit the proliferation,invasion and induced apoptosis of esophageal carcinoma cell line TE-1.The experimental results show that IGFBP-2 may play an important role in the etiopathogenesis of esophageal cancer.IGFBP-2 may be a target gene for esophageal cancer treatment.
