慢性不完全性睡眠剥夺大鼠对海马、前脑皮质NG及血清TGF-β1表达的影响

Effects of Chronic Incomplete Sleep Deprivation on Expression of Hippocampal and Anterior Cortex NG and Serum TGF-β1 in Rats

摘要 为探讨慢性疲劳大鼠睡眠剥夺后对血清转化生长因子(TGF-β1)、海马、前脑皮质NG含量的影响,采用负重力竭游泳法制备CFS大鼠模型,用小平台水环境法剥夺睡眠12 h,连续应激21 d后,Morris水迷宫测量大鼠学习记忆能力,ELISA检测血清TGF-β1的含量,Western blot法检测海马、前脑皮质NG含量,LDH释放法检测脾脏NK细胞活性。结果表明: 与对照组相比,睡眠剥夺组,Morris水迷空间搜索实验,总路程、平均速度显著提高(P<0.05),经过平台次数降低(P<0.05);血清TGF-β1和海马NG含量升高(P<0.05);前脑皮质NG含量降低(P<0.05);脾脏NK细胞活性降低(P<0.05)。说明TGF-β1抑制NK细胞活性使机体免疫功能低下可能是CFS发生的重要病理机制之一。 To explore the effects of sleep deprivation on serum transformed growth factor (TGF-β1),NG in hippocampus and forebrain cortex.CFS rat model was prepared by using the method of weight-bearing exhaustion swimming,and sleep was deprived for 12 hours with the method of small platform water environment.20 SD rats were randomly divided into the control group and the sleep deprivation group.Serum TGF-β1 was detected by ELISA.Western blot was used to detect the content of NG in hippocampus and forebrain cortex.Activity of spleen NK cells was detected by LDH release.The results of the Morris water maze search experiment show that,compared with the control group,the total distance and average speed of the sleep deprivation group are improved (P<0.05) and the number passes through the platforms is decreased (P<0.05).Serum TGF-β1 and NG in hippocampus content were increased(P<0.05).NG content in the forebrain cortex was decreased (P<0.05);The activity of NK cells in the spleen was decreased (P<0.05).TGF-β1 inhibits NK cell activity and makes the body's immune function low may be one of the important pathological mechanisms of CFS.
