PVT1和miR-195在甲状腺乳头状癌组织中的表达及临床意义 被引量:1

摘要 目的:探讨长链非编码RNA浆细胞瘤多样异位基因1(lncRNA PVT1)和miR-195在甲状腺乳头状癌(PTC)组织中的表达及临床意义。方法:选取2017年1月至2018年12月本院内分泌科及甲状腺外科收治的120例PTC患者进行研究(PTC组),120例甲状腺良性病变患者作为良性病变组,癌旁>4 cm以外的正常甲状腺组织为对照组,采用荧光定量PCR(qRT-PCR)检测组织中PVT1和miR-195水平,与临床病理参数关系,分析二者表达相关性及对PTC诊断价值。结果:PTC组患者癌组织中PVT1水平高于良性病变组、对照组(P<0.05),良性病变组与对照组比较差异无统计学意义(P>0.05),三组间比较差异有统计学意义(P<0.05);PTC组患者癌组织中miR-195水平低于良性病变组、对照组(P<0.05),良性病变组低于对照组(P<0.05),三组间比较差异有统计学意义(P<0.05);PVT1、miR-195表达水平与患者性别、年龄、肿瘤大小无关(P>0.05),与TNM分期、淋巴结转移有关(P<0.05);通过Pearson相关性分析显示,PTC患者组织中PVT1和miR-195表达呈负相关性(r=-0.840,P=0.000)。ROC曲线显示,PVT1的AUC为0.769,敏感性为60.00%,特异性为90.00%,miR-195的AUC为0.892,敏感性为81.67%,特异性为83.33%。结论:PTC患者组织中PVT1高表达,miR-195低表达,二者表达呈负相关性,与TNM分期、淋巴结转移有关,具有较高诊断价值,可能参与PTC发生、发展。 To investigate the expressions and clinical significances of long non-coding RNA plasmacytoma variant translocation 1(lncRNA PVT1)and miR-195 in thyroid papillary carcinoma(PTC)tissues.Methods:120 patients with PTC admitted to the department of endocrinology and thyroid surgery of our hospital from January 2017 to December 2018 were selected for the study(PTC group),120 patients with benign thyroid lesions as the benign lesion group,and normal thyroid tissues beyond 4 cm adjacent to cancer as the control group.Quantitative fluorescence PCR(qRT-PCR)was used to detect the levels of PVT1 and miR-195 in tissues,and their relationships with clinicopathological parameters.The correlation between the two expressions and their diagnostic values of PTC were analyzed.Results:The level of PVT1 in cancer tissues of PTC group was higher than that of benign lesion group and control group(P<0.05),there was no significant difference between benign lesion group and control group(P>0.05),there were significant differences among the three groups(P<0.05).The level of miR-195 in cancer tissues of PTC group was lower than that of benign lesion group and control group(P<0.05).The benign lesion group was lower than the control group(P<0.05).There were significant differences among the three groups(P<0.05).The expression levels of PVT1 and miR-195 were not correlated with gender,age,and tumor size(P>0.05),but with TNM stage and lymph node metastasis(P<0.05).Pearson correlation analysis showed that the expressions of PVT1 and miR-195 were negatively correlated in PTC patients(r=-0.840,P=0.000).ROC curve showed that the AUC of PVT1 was 0.769,the sensitivity was 60.00%,and the specificity was 90.00%,the AUC of miR-195 was 0.892,the sensitivity was 81.67%,and the specificity was 83.33%.Conclusion:The expression of PVT1 is high and the expression of miR-195 is low in PTC patients.They are negatively correlated,and related to TNM and lymph node metastasis.It has high diagnostic value and may be involved in the occurrence and development of PTC.
