Simultaneous Determination of Three Constituents and Characteristic Spectrum of Bran Fructus Aurantii Granules by HPLC

摘要 目的:建立能够便捷同步测定麸炒枳壳配方颗粒的特征图谱并实现对其主要成分柚皮苷、橙皮苷以及新橙皮苷进行含量测定的方法,从定性和定量两个角度实现客观评价麸炒枳壳配方颗粒的质量。方法:采用HPLC,色谱柱为C18,以乙腈和pH为3的磷酸水溶液作为流动相进行梯度洗脱,检测波长设定283 nm,流速为1 mL·min^-1,并对10批麸炒枳壳配方颗粒的相关指标进行检测。结果:特征图谱检测结果共呈现5个特征峰。含量测定结果表明,柚皮苷、橙皮苷、新橙皮苷分别在0.038 8~1.553 0、0.001 9~0.077 4、0.021 2~0.850 2 μg线性关系良好,r>0.999 8,3种成分平均回收率分别为98.86%、99.66%、99.21%,RSD分别为2.11%、1.52%、2.21%。结论:该方法操作简便,准确,重复性好,可作为麸炒枳壳配方颗粒进行质量研究和评价的依据。 Objective:To establish a method for determining the contents of narigin,hesperidin,neohesperidin and characteristic spectrum in Bran Fructus Aurantii Granules by HPLC.It was developed and validated for quality evaluation for Bran Fructus Aurantii Granules.Methods:The chromatography column was C18 column.The mobile phase consisted of acetonitrile and water(adjusting pH to 3 with phosphoric acid)with the gradient mode at a flow rate of 1 mL·min^-1,wavelength set at 283 nm.Ten batches of Bran Fructus Aurantii Granules were measured.Results:The Characteristic spectrum showed five characteristic peaks.The results of content determination showed that the linear relationship of narigin,hesperidin and neohesperdin were good at 0.038 8-1.533 0,0.001 9-0.077 4,0.021 2-0.850 2 μg,r>0.9998,the average recovery were 98.86%,99.66%,99.21%,RSD were 2.11%,1.52%,2.21%,respectively.Conclusion:A method for simultaneous determination of the characteristic spectrum and three contents of Bran Fructus Aurantii Granules was established.The method is accurate,stable and credible.It can be used for the quality study and evaluation of Bran Fructus Aurantii Granules.
