摘要 目的探究表没食子儿茶素没食子酸酯(EGCG)对人宫颈癌细胞株Hela细胞生物学行为及血管内皮生长因子(VEGF)、基质金属蛋白酶9(MMP-9)的表达的影响。方法取人宫颈癌Hela细胞进行培养分别给予0μmol、10μmol、20μmol、30μmol、40μmol EGCG进行处理。采用CCK8法、流式细胞法、Transwell法分别检测不同剂量EGCG处理下细胞增殖、凋亡、侵袭力;采用RT-PCR、Western Blot法检测细胞VEGF、MMP-9基因表达情况。结果①EGCG处理后,Hela细胞增殖抑制率、凋亡率升高,侵袭细胞数减少(P <0.05);随着EGCG剂量的升高,Hela细胞增殖抑制率、凋亡率、侵袭细胞数升高或降低幅度升高(P <0.05)。②未经EGCG(处理浓度=0)组别Hela细胞VEGF、MMP-9基因mRNA、蛋白呈高表达,处理组别细胞VEGF、MMP-9基因mRNA、蛋白表达量降低,随着处理浓度的升高,其表达量降低幅度更大(P <0.05)。结论 EGCG可呈剂量依赖性抑制人宫颈癌Hela细胞的增殖及侵袭,促使其凋亡,对于恶性生物学行为有较好的抑制效果,VEGF、MMP-9等基因表达的抑制可能是其机制之一。 Objective To explore effects of epigallocatechin-3-gallate(EGCG) on biological behaviors of human cervical cancer cell line Hela cells and expression of vascular endothelial growth factor(VEGF) and matrix metalloproteinase-9(MMP9).Methods Human cervical cancer Hela cells were cultured and treated with 0μmol,10μmol,20μmol,30μmol,40μmol EGCG.CCK8,flow cytometry and Transwell were applied to detect cell proliferation,apoptosis and invasion ability under different-dose EGCG treatment.The expression of VEGF and MMP-9 genes was detected by RT-PCR and Western Blot.Results①After EGCG treatment,proliferation inhibition rate and apoptosis rate of Hela cells were increased,while number of invasive cells was decreased(P <0.05).With increase of EGCG dose,proliferation inhibition rate,apoptosis rate and increase or decrease amplitude of invasive cells number were increased(P <0.05).②In group without EGCG treatment(treatment concentration=0),mRNA and protein of VEGF and MMP-9 genes in Hela cells were highly expressed.The expression quantities of VEGF and MMP-9 mRNA and protein were decreased in treatment group(P <0.05).With increase of treatment concentration,decrease amplitude of their expression quantities is more significant(P <0.05).Conclusion EGCG can inhibit proliferation and invasion of human cervical cancer Hela cells,promote their apoptosis,which is of good inhibitory effect on malignant biological behaviors.The inhibition of VEGF,MMP-9 and other gene expression may be one of the mechanisms.
