摘要
为了建立红糖的高效液相(HPLC-DAD)指纹图谱的质量评价方法。采用MYCOTOXTM反相C18色谱柱(4.6mm×250 mm,5μm),以0.1 mol/L磷酸二氢钠(pH 2.3)-乙腈为流动相,等度洗脱,流速为0.5mL/min,柱温30℃,进样量20μL,检测波长210nm,根据相似度评价,结合化学计量学方法对红糖进行质量评价。结果建立了21批红糖的HPLC-DAD指纹图谱,标定共有峰13个,其中19批样品指纹图谱的相似度均大于0.913,通过聚类分析可将样品聚为2类,结合主成分分析、正交偏最小二乘法-判别分析发现其中11个成分是造成不同批次样品差异性的主要标记物。所建立的液相指纹图谱特征性和专属性强,可作为红糖内在质量控制的有效方法。
The present study is to establish the fingerprint spectra for the quality evaluation of brown sugar by HPLC-DAD.The chromatographic conditions are defined as MYCOTOXTM reversed-phase C18(4.6 mm×250 mm,5μm).The mobile phase is acetonitrile-0.1 mol/L monosodium phosphate(pH 2.3)in isocratic elution,and the flow rate is 0.5 mL/min.The column temperature is 30℃,the injection volume is 20μL,the detection wavelength is 210 nm.According to the similarity evaluation,the chemometrics method is used to evaluate the quality of brown sugar.The fingerprint spectra of 21 batches of brown sugar are established,there are 13 common peaks in the fingerprint spectra.The similarity of fingerprint spectra of 19 batches of samples is greater than 0.913.The samples are classified into two groups by hierarchical cluster analysis combined with principal component analysis(PCA)and orthogonal partial least squares-discriminant analysis(OPLS-DA),and eleven components are the main markers that cause differences in the different batches of samples.The established HPLC-DAD fingerprint spectra are highly specific,and can be used as an effective method for internal quality control of brown sugar.
