摘要
目的:探讨脂肪间充质干细胞(ADSCs)外泌体对心肌梗死中心肌细胞的保护作用。方法:从SD大鼠中分离、培养ADSCs并提取外泌体,用透射电镜和免疫印迹方法进行鉴定;建立体外无血清无氧培养的心肌梗死细胞模型,在外泌体处理后用TUNEL和CCK8法分别检测H9C2细胞的凋亡和增殖变化。结果:ADSCs的外泌体直径为30~120 nm,呈圆形或椭圆形,表达CD9、CD63、CD81等外泌体标记蛋白。在外泌体处理48 h后,H9C2细胞的凋亡明显减少,增殖明显增多。结论:ADSCs外泌体在心肌梗死中可保护心肌细胞,并减少其凋亡,可作为治疗心肌梗死的新方法。
Objective:To explore the effect of exosomes secreted from adipose tissue-derived stromal cells(ADSCs)on myocardial infarction.Methods:Exosomes secreted from ADSCs were identified by protein marker expression and transmission electron microscopy.A cell model of myocardial infarction was established through serum-free and anaerobic culture.The apoptosis and proliferation of H9C2 cells were detected by TUNEL and CCK8 after exosome treatment.Results:Electron micrographs showed that ADSCs-derived exosomes exhibited round or oval structures with a size of approximately 30–120 nm.Western blotting analysis confirmed that exosome expressed CD9,CD63 and CD81 exosome markers.The apoptosis of H9C2 cells was significantly reduced and the proliferation increased after 48 h exosome treatment.Conclusion:ADSCs-derived exosomes could protect myocardial cells from apoptosis in myocardial infarction,and could be used as a new method to treat myocardial infarction.
