摘要
目的探讨线粒体核糖体蛋白S5(MRPS5)在肾透明细胞癌(ccRCC)组织中的表达及其与患者预后的关系。方法采用免疫组织化学和免疫印迹技术检测ccRCC患者新鲜癌组织和癌旁组织中MRPS5的相对表达;采用免疫组织化学技术检测ccRCC患者石蜡包埋癌组织标本中MRPS5的表达;χ2检验分析MRPS5表达水平与患者临床病理参数的关系;Kaplan-Meier法描绘生存曲线,Log-rank检验分析MRPS5表达水平与患者生存状况的关系;采用Cox比例风险回归模型分析影响患者预后的临床因素。结果与癌旁组织相比,癌组织中MRPS5的表达水平均明显较低;在104例ccRCC患者癌组织中,64例(61.5%)高表达MRPS5,40例(38.5%)低表达MRPS5;MRPS5表达水平与肿瘤T分期(P=0.024)及肿瘤坏死(P=0.044)显著相关;MRPS5低表达组的患者总生存期(OS)和无病生存期(DFS)均显著短于高表达组(P<0.05);单因素Cox回归分析发现,ccRCC患者OS和DFS均与肿瘤大小、T分期、N分期、TNM临床分期、Fuhrman分级、肿瘤坏死及MRPS5表达水平等多种因素相关(P<0.05);多因素Cox回归分析发现,ccRCC患者OS与DFS均和N分期、Fuhrman分级及MRPS5表达水平相关(P<0.05)。结论MRPS5在ccRCC患者的癌组织中的表达低于癌旁组织,MRPS5低表达是影响患者预后的独立危险因素,有望成为判断患者预后的一种重要生物标志物及新的治疗靶点。
Objective To investigate the expression of mitochondrial ribosomal protein S5(MRPS5)in the tissues of clear cell renal cell carcinoma(ccRCC)and its relationship with the prognosis of patients.Methods MRPS5 expression in fresh cancerous tissues and adjacent normal tissues of patients with ccRCC were detected by immunohistochemistry and immunoblotting.MRPS5 expression in paraffin-embedding cancer tissues of specimens with ccRCC was assayed by immunohistochemistry.The relationship between MRPS5 expression level and clinical pathological parameters was analyzed byχ2 test.The survival curve was plotted by Kaplan-Meier,and the relationship between MRPS5 expression level and patient survival status was tested by Log-rank.Cox proportional risk regression model was used to analyze the clinical factors influencing the prognosis of patients.Results The expression of MRPS5 in cancerous tissues was remarkably lower than that in adjacent normal tissues.In the 104 cases of cancerous tissues,there were 64 patients(61.5%)with MRPS5 high expression and 40 patients(38.5%)with MRPS5 low expression.In addition,MRPS5 expression levels were significantly associated with T classification(P=0.024)and tumor necrosis(P=0.044).Moreover,the overall survival(OS)and disease free survival(DFS)of patients with low MRPS5 expression levels were dramatically shorter than those of patients with high MRPS5 expression levels.Univariate Cox regression analysis showed that OS and DFS were correlated with tumor size,T stage,N stage,TNM clinical stage,Fuhrman grade,tumor necrosis and MRPS5 expression level(P<0.05).Multivariate Cox regression analysis found that OS and DFS were correlated with N stage,Fuhrman grade and MRPS5 expression level in ccRCC patients(P<0.05).Conclusion MRPS5 expression in cancerous tissues is lower than that in the adjacent normal tissues of ccRCC patients.Low MRPS5 expression is an independent risk factor affecting the prognosis of patients.Therefore,it is expected to be an important prognosis biomarker and a new therapeutic target for patients with ccRCC.
