破壁灵芝孢子粉联合他莫昔芬对人乳腺癌MCF-7细胞皮下移植瘤生长增殖的影响 被引量:4

摘要 目的观察破壁灵芝孢子粉联合他莫昔芬对人乳腺癌MCF-7细胞皮下移植瘤生长增殖的影响。方法建立人乳腺癌MCF-7细胞裸鼠移植模型,把44只成瘤裸鼠随机分成4组,分别为生理盐水组(对照组)、破壁灵芝孢子粉组、他莫昔芬组和联合组(破壁灵芝孢子粉+他莫昔芬)。用药21d,记录用药后第3、6、10、13、17、21d瘤体体积并计算其抑制率。将瘤体剥离后,分别进行HE染色对比分析及ER-α、ER-β、P53及Ki-67四种免疫组化分析。结果与对照组相比,破壁灵芝孢子粉组、他莫昔芬组及联合组的移植瘤重量体积均显著减少(P<0.05),他莫昔芬组、破壁灵芝孢子粉组及联合组的抑瘤率分别为57.3%、56.4%及69.3%。从免疫组化分析来看,P53蛋白表达在联合组下调明显,低于其他三组,且具有统计学差异(P<0.05),而ER-α、ER-β及Ki-67三种免疫组化指标表达率在对照组、破壁灵芝孢子粉组、他莫昔芬组及联合组内的差异不大,无统计学差异(P>0.05)。结论破壁灵芝孢子粉联合他莫昔芬能够抑制肿瘤细胞增长及繁殖的作用,并显著降低肿瘤P53的表达达到潜在的抗肿瘤转移作用。 Objective To observe the effect of spore powder of broken ganoderma lucidum combined with tamoxifen on the growth and proliferation of subcutaneous transplanted human breast cancer MCF-7 cells.Methods To establish a nude mouse transplantation model of human breast cancer MCF-7 cells,twenty-eight tumorigenic nude mice were divided into four groups on average,they were saline group(control group),spore powder group,tamoxifen group and combination group(spore powder+tamoxifen group).Tumor volume was recorded on the 2nd,4th,6th,8th,10th,12th day after administration and the inhibition rate was calculated.After the tumor was dissected,HE staining,ER-α,ER-β,P53 and Ki-67 immunohistochemistry were performed.Results Compared with the control group,the weight and volume of transplanted tumors in spore powder group,tamoxifen group and combination group were significantly reduced(P<0.05),the inhibition rates of spore powder group,tamoxifen and combination groups were 57.3%,56.4%and 69.3%respectively.From the immunohistochemical analysis,the expression of P53 protein in the combined group was significantly lower than that in the other three groups,with statistical significance(P<0.05),while the expression rates of ER-alpha,ER-beta and Ki-67 were not significantly different in the control group,the broken Ganoderma lucidum spore powder group,tamoxifen group and the combined group(P>0.05).Conclusion Spore powder of broken ganoderma lucidum combined with tamoxifen can inhibit the growth and proliferation of tumor cells.
