大鼠梭形动脉瘤模型的制作研究

摘要 目的探讨采用猪胰弹性蛋白酶消化大鼠右侧颈总动脉制作梭形动脉瘤模型的可行性。方法应用猪胰弹性蛋白酶消化大鼠右侧颈总动脉建立动脉瘤模型,于7d、14d行DSA检测动脉瘤直径,即刻灌注取标本行HE染色、免疫组化等研究观察该模型的影像学和病理学变化。结果动脉血管造影显示正常对照组大鼠右侧颈总动脉直径(0.68±0.04mm),生理盐水对照组大鼠右侧颈总动脉直径(0.69±0.03mm),实验A组7d梭形动脉瘤宽径(1.37±0.02mm),实验B组14d梭形动脉瘤宽径(1.39±0.03mm)。实验组A和B造影的动脉瘤宽径分别与对照组的比较运用配对样本T检验P<0.05,差异有统计学意义。实验组A造影的动脉瘤宽径与实验组B造影的动脉瘤宽径相互比较运用配对样本T检验P>0.05,差异无统计学意义。病理学观察发现实验组动脉瘤外膜破坏,弹力层在动脉瘤颈处部分消失。对照组显示血管壁结构完整。结论利用外科手术方法和弹性蛋白酶来消化部分动脉血管壁,制作和病理学基本相似的梭形动脉瘤模型。 Objective Use of porcine pancreatic elastase production of carotid artery fusiform aneurysm model in rats.Methods An experimental aneurysm model was created in rats by means of porcine pancreatic elastase digestion the origin of right common carotid artery(RCCA).The animals were sacrificed at 7 days,14 days,and Immunohistochemistry,HE staining,digital subtraction angiography(DSA)evaluation of the aneurysm was performed.Results Arterial angiography showed the diameter of the right common carotid artery in the control group(0.68±0.04 mm),Normal saline control group rats right common carotid artery diameter(0.69±0.03 mm),Experimental group A 7 days aneurysm wide diameter(1.37±0.02 mm),Experimental group B 14 days aneurysm wide diameter(1.39±0.03 mm).Experimental group A and B angiography aneurysms wide diameter compared with the control group using paired samples T-test P<0.05,The difference was statistically significant.Experimental group A angiography aneurysms wide diameter and experimental group B angiography aneurysms wide diameter compared with each other using paired samples T-test P>0.05,The difference was not statistically significant.Pathologic analysis found that the experimental group aneurysms outer membrane damage,Elastic layer of the neck of the aneurysm partial disappearance.Control group showed vascular wall structure integrity.Conclusion Surgical methods and elastase were used to digest part of the arterial wall,Fusiform aneurysm models which are similar to human intracranial aneurysms in morphology and pathology can be made.
