摘要
目的肝星状细胞(HSC)的激活受多条信号通路的调节,是肝内型门脉高压发生的关键事件。此研究对静态和活化HSC二代测序结果进行了全面的生物信息学分析,旨在深入了解HSC激活的关键基因及其信号通路。方法通过基因功能分析——功能的显著性分析和KEGG信号通路对差异表达信使RNA(mRNA)进行分析。结果根据基因功能分布发现,HSC转型过程中主要涉及细胞凋亡、增殖、迁移等40种功能,KEGG信号通路提示30条信号通路富集度高。结论在HSC活化向肌成纤维母细胞分化的过程中存在mRNA基因功能和信号通路的差异调控,这为肝纤维化及门脉高压的发病机制和诊治研究提供了新方向。
Objective The activation of hepatic stellate cells(HSC)is regulated by multiple signaling pathways,which is a key event in the development of intrahepatic portal hypertension.This paper intends to gain an in-depth understanding of key genes and their signaling pathways activated by HSC.Methods Differentially expressed mRNAs were analyzed by gene function analysis--GO analysis and KEGG signaling pathways.Results According to the distribution of gene function,40 kinds of functions such as apoptosis,proliferation and cell migration were mainly involved in the process of HSC transformation.KEGG suggests that 30 pathways were highly enriched.Conclusion During the differentiation of HSC activation into myofibroblasts,there are differential regulation of mRNA gene functions and signaling pathways,which provides a new pathway for the pathogenesis and diagnosis of liver fibrosis and portal hypertension.
