瑶山鸡TRH基因编码区序列变异及其与繁殖性状的相关性 被引量:2

摘要 为明确促甲状腺激素释放激素(thyrotropin-releasing hormone,TRH)作为鸡繁殖性状辅助选育基因的可行性,采用PCR产物测序法对瑶山鸡TRH基因编码区进行序列变异检测,并与繁殖性状进行关联分析。结果表明:瑶山鸡TRH基因存在1个编码区变异位点、1个外显子非编码区变异位点和1个内含子区变异位点,编码区变异位点引起蛋白质相应氨基酸发生错义突变。外显子区A2699T、G3857T变异位点与300日龄产蛋量、首次就巢日龄、首次就巢持续时间、300日龄就巢次数存在显著或极显著关联。外显子区A2699T、G3857T位点可作为瑶山鸡繁殖性状选育的辅助选择标记。 In order to clarify the feasibility of thyrotropin-releasing hormone(TRH)as an auxiliary breeding gene for chicken breeding traits,the PCR product sequencing method was employed to detect sequence mutations in the coding region of the TRH gene of Yaoshan Chicken,and to conduct the correlation analysis of breeding traits.Results:There was one mutation site in coding region,one mutation site in exon non-coding region and one mutation site in intron region in the TRH gene of the Yaoshan Chicken.The coding region mutation sites gave rise to the missense mutation of the corresponding amino acid of the protein.The exon A2699T and G3857T mutation loci were significantly or extremely significantly correlated with egg production at 300 d age,first nesting day,first nesting duration,and nesting times at 300 d age.Exon regions A2699T and G3857T can be used as auxiliary selection markers for breeding traits of Yaoshan Chicken.
