摘要
为西伯利亚鲟(Acipenser baerii)精子超低温冷冻保存体系优化及冷冻精子保存效果提供理论支撑,以西伯利亚鲟新鲜精子为对照,研究超低温冷冻保存(-196℃)条件下精子中总ATP酶(ATPase)、谷胱甘肽还原酶(GR)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、琥珀酸脱氢酶(SDH)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)等的酶活性变化。结果表明:超低温冷冻前后西伯利亚鲟精子内的GR酶活性从(23.36±1.69)U/mL升至(94.61±4.13)U/mL,差异显著;CK酶、LDH酶、SDH酶、CAT酶及SOD酶的酶活性均显著降低,分别从(6.64±0.58)U/mL、(6695.78±123.47)U/L、(24.91±1.63)U/mL、(112.33±6.38)U/mL、(61.99±4.15)U/mL降至(5.27±0.23)U/mL、(4388.32±93.28)U/L、(17.45±0.94)U/L、(67.58±3.87)U/mL、(46.38±2.54)U/mL;总ATP酶从冻前的(18.77±1.23)U/mL降至(16.32±0.87)U/mL,差异不显著。超低温冷冻保存对西伯利亚鲟精子酶活性均有显著影响。
The effect of cryopreservation(-196℃)on ATP,GR,CK,LDH,SDH,CAT and SOD activity of Acipenser baerii spermatozoa was studied by taking fresh A.baerii spermatozoa as CK to provide the theoretical support for cryopreservation system optimization and frozen preservation effect of A.baerii spermatozoa.Result:The GR activity of A.baerii spermatozoa rises from(23.36±1.69)U/mL(before cryopreservation)to(94.61±4.13)U/mL(after cryopreservation)significantly.The CK,LDH,SDH,CAT and SOD activity of A.baerii spermatozoa significantly decreases from(6.64±0.58)U/mL,(6695.78±123.47)U/L,(24.91±1.63)U/mL,(112.33±6.38)U/mL and(61.99±4.15)U/mL(before cryopreservation)to(5.27±0.23)U/mL,(4388.32±93.28)U/L,(17.45±0.94)U/L,(67.58±3.87)U/mL and(46.38±2.54)U/mL(after cryopreservation)respectively.The ATP activity of A.baerii spermatozoa decreases from(18.77±1.23)U/mL(before cryopreservation)to(16.32±0.87)U/mL(after cryopreservation)but the difference is not significance.In conclusion,cryopreservation has the significant effect on enzymatic activity of A.baerii spermatozoa.
