重症急性胰腺炎继发感染早期肝损伤时冷诱导RNA结合蛋白的表达和意义 被引量:1

The Role of Cold-Inducible RNA-Binding Protein (CIRP) in Early Liver Damage

摘要 目的:研究冷诱导RNA结合蛋白(CIRP)在重症急性胰腺炎(SAP)和重症胰腺炎继发感染(SISAP)早期肝损伤中的表达、意义和可能机制。方法:30只雄性SD大鼠随机分为对照组、重症急性胰腺炎组(SAP组)、重症急性胰腺炎继发感染组(SISAP组)。通过腹腔注射精氨酸、大肠杆菌构建动物模型,造模后24 h取部分胰腺及肝组织光学取实时聚合酶链反应和Western blotting方法检测肝组织中CIRP mRNA和蛋白的表达,光学显微镜下进行组织病理学评分,同时血清学测定血淀粉酶、谷氨酸转氨酶(ALT)、肿瘤坏死因子(TNF)-α、C反应蛋白(CRP)及CIRP表达变化。结果:与对照组比较,SAP组和SISAP组胰腺及肝脏病理评分、血清淀粉酶、ALT明显升高(P<0.05),SISAP组更显著(与SAP组比较,P<0.05)。与对照组比较,SAP组和SISAP组的血清CRP、TNF-a均显著升高(P<0.05),但SAP组和SISAP组间比较,差异无统计学意义(P>0.05)。与对照组(1.01±0.17)相比,SAP组肝脏组织CIRP mRNA的表达明显升高(P<0.05),SISAP组增高更显著(P<0.05)。CIRP蛋白表达水平与mRNA表达的趋势一致,并且CIRP水平与肝功能指标ALT,肝损伤病理学评分结果相一致。结论:CIRP可能参与SISAP引起的早期肝损伤的病理进程。 [Objictive]To explore the role and the mechanism of cold-inducible RNA-binding protein triggering receptor(CIRP)in early liver damage of severe acute pancreatitis with secondary infection.[Method]Thirty male SD rats were randomly divided into the control group,the severe acute pancreatitis(SAP)group and the secondary infection of SAP(SISAP)group.The animal model was established by intraperitoneal injection of L-arginine and E.coli.After 24 hours,Part of the pancreas and liver tissue were harvested for histopathological score under microscope.The expression of CIRP at mRNA and protein level in liver tissue was detected through Real-time PCR and Western Blot.Meanwhile,the serum 1evels of amylase,glutamate aminotransferase(ALT),CIRP,tumor necrosis factor(TNF)-α,and C-reactive protein(CRP)were determined by automatic biochemical analyzer.[Result]Compared with the control group,the histological score of pancreas and liver,serum amylase,ALT increased in all the SAP groups,especially in SISAP groups(P<0.05).The CRP and TNF-a expression were increased statistically in SAP and SISAP groups as compared with those of Sham group,however,there was no significant difference between the SAP and SISAP groups(P>0.05).Compared with the control group(1.01±0.17),The expression of CIRP mRNA in liver tissue of SAP group was significantly higher(P<0.05),The expression of in SISAP group was significantly highest(P<0.05).The expression of CIRP at protein level was same as mRNA expression,and the level of CIRP was consistent with the pathological score of liver injury in ALT AST,the index of liver funtion.[Conclusion]CIRP may play an important role in the pathogenesis of liver damage caused by severe acute pancreatitis.
