Effects of formin homology protein 2 on the biological function of human lung cancer HCC827 cells

摘要 目的探讨同源形成素蛋白2(formin homology proteins,FMNL2)在肺癌细胞系中的表达,及其与肺癌细胞侵袭、迁移的相关性。方法通过细胞转染siRNA降低FMNL2的表达,采用细胞增殖实验、划痕实验和侵袭实验等观察FMNL2对肺癌HCC827细胞的影响。结果敲低FMNL2的表达,发现肺癌HCC827细胞的活力以及增殖能力均降低,而在划痕实验和侵袭实验中,敲低FMNL2也降低了划痕的愈合速度和侵袭细胞数量。结论FMNL2的高表达能够促进肿瘤的恶性生物学行为,FMNL2参与并调控了肺癌细胞的发生和发展过程,并可能成为影响患者预后的关键因子。 Objective To study the expression of FMNL2 in the lung cancer cell lines and its relations with invasion and metastasis of human lung cancer cells. Methods The expression of FMNL2 was decreased by transfecting siRNA into lung cancer cells. The effect of FMNL2 on the lung cancer HCC827 cells was detected by using MTT assay, wound healing and transwell assay. Results The viability as well as the proliferation of lung cancer HCC827 cells were reduced by inhibiting FMNL2 expression, while knockdown of FMNL2 also reduced the healing rate of scratches and invasive cell number in scratch assay and invasion assay. Conclusion High expression of FMNL2 may promote the malignant biological behavior of tumor. FMNL2 participates in and regulates the development and progression of lung cancer cells and may become a key factor affecting the prognosis of patients.
