高效液相色谱法测定葛仙米水溶性多糖含量 被引量:2

摘要 通过建立高效液相法(HPLC)测定葛仙米水溶性多糖含量的方法,对HPLC测定葛仙米水溶性多糖含量的方法学进行考察。以纯化的葛仙米多糖为标准品,应用HPLC对经过不同预处理的葛仙米水溶性多糖进行含量测定。结果表明:选择柱型为TSKgel G6000 PWXL液相色谱分析柱的效果较好,标准品的定量上、下限分别为10mg/mL和0.078625mg/mL;标准溶液浓度(Y)在0.15625~5mg/mL范围内,峰面积与进样量(X)呈良好的线性关系,线性方程Y=1995695.548X-170.597、R^2=0.999;精密度、重复性和稳定性的RSD分别为1.59%、1.13%、2.56%,平均加样回收率为97.8%、RSD为1.72%,说明该方法灵敏度高、重复性和稳定性较好,试验结果可靠,所建方法适用于葛仙米水溶性多糖含量的测定。 A HPLC(High Performance Liquid Chromatography)method for the determination of Nostoc sphaeroids kiitzing water-soluble polysaccharide content was developed.Using the purified Nostoc sphaeroids kiitzing water-soluble polysaccharide as standard,the content of water-soluble polysaccharides from Nostoc sphaeroids kiitzing after different pretreatment was determined by HPLC.The results indicated that the analytical column of TSKgel G6000 PWXL is better.The standard upper and lower bound were 10 mg/mL and 0.078625 mg/mL,the linear equation was F=1995695.548X-170.597,R^2=0.999;The precision,rrepeatability and stability of the relative standard deviation(RSD)were 1.59%,1.13%and 2.56%,the average recovery rate was 97.8%and RSD was 1.72%,indicating that the method has high sensitivity,repeatability and stability.Thus HPLC method is suitable for determining Nostoc sphaeroids kiitzing water-soluble polysaccharide content.
