摘要 目的探究细胞分裂周期蛋白25同源蛋白C(CDC25C)在肝癌中的表达及其意义,为指导临床治疗、评估患者预后及制订肝癌防治策略与措施提供科学依据。方法利用基因表达谱数据交互分析(GEPIA)数据库、UALCAN在线数据库分析CDC25C在肝癌组织中的差异表达,以及CDC25C在肝癌组织中相应表达水平与预后的关系。应用癌症基因组图谱(TCGA)数据集分析CDC25C在肝癌及其他组织中的基因突变情况。通过相互作用基因库检索工具(STRING)数据库进一步构建蛋白质-蛋白质相互作用(PPI)网络。结果与正常肝组织样品相比,肝癌组织中CDC25C的表达明显增强(P<0.01)。CDC25C的高表达可明显降低肝癌患者的无病生存期和总体生存期(P<0.01)。CDC25C在肝癌组织中的表达与肝癌的分级、分期相关(P<0.05或P<0.01)。另外,CDC25C的4个特异性基因突变(V41A、L87H、N222K和X309-splice)发生在肝癌样品中,这些独特的突变在其他肿瘤组织中均未发现。GO基因功能注释及KEGG通路富集分析结果提示,CDC25C可能与细胞周期信号通路密切相关。结论CDC25C在一定程度上影响着肝癌的发生、发展,有望成为肝癌基因诊断、治疗及预后的靶点。 Objective To explore the expression and significance of cell division cycle 25 homologus protein C(CDC25C)in liver cancer,in order to provide scientific basis for guiding clinical treatment,assessing the prognosis of patients,and formulating strategies and measures for liver cancer prevention.Methods The differential expression of CDC25C in liver cancer tissues and the relationship between the corresponding expression level of CDC25C in liver cancer tissues and prognosis were analysed by using the gene expression profiling interactive analysis(GEPIA)database and UALCAN online database.The Cancer Genome Atlas(TCGA)data set was used to analyze the mutations of CDC25C in liver cancer and other tissues.The protein-protein interaction(PPI)network was further constructed by using the search tool for the retrieval of interacting genes(STRING)database.Results Compared with normal liver tissues,the expression of CDC25C in liver cancer tissues was significantly enhanced(P<0.01).A higher expression of CDC25C resulted in a remarkably shorter disease free survival as well as overall survival(P<0.01).The expression of CDC25C in liver cancer tissues was related to the grade and stage of liver cancer(P<0.05 or P<0.01).Additionally,four specific gene mutations of CDC25C,including V41A,L87H,N222K and X309-splice,occurred in liver cancer samples,and these unique mutations were not found in any other tumor tissues.Finally,the results of GO function annotation and KEGG pathway enrichment analysis showed that CDC25C might be associated with cell cycle signaling pathway.Conclusion To some extent,CDC25C affects the occurrence and development of liver cancer,and is expected to be a biomarker for gene diagnosis,treatment and prognosis of liver cancer.
