比格犬慢性冬眠心肌模型的制备 被引量:1

摘要 目的制备可供实验使用的比格犬慢性冬眠心肌模型。方法选取16只健康成年比格犬,分为A组(冬眠心肌组,8只)和B组(假手术组,8只)。A组采用开胸丝线结扎法造成左前降支狭窄(75%),术后每日保持室外活动4 h,靶血管供血区域慢性缺血,制备慢性冬眠心肌模型。B组将丝线穿过前降支下方,但不结扎,其余同A组。两组均于术后3个月终止实验。采用超声心动图多巴酚丁胺负荷试验观察A组左心室壁运动,采用超声心动图观测两组收缩期室壁增厚率(WT%)、左心射血分数(LVEF)等指标。结果A组左心室壁运动异常,WT%、LVEF减低。结论开胸丝线结扎,术后辅以规律室外运动的方法制备犬冬眠心肌模型,模型制备方法简单,成功率高,对实验人员技术要求低,实验安全无射线,是一种简单可行的冬眠心肌模型的制备方法。 Objective To establish a beagle dog chronic hibernating myocardium model for experimental use.Methods Sixteen healthy adult beagles were divided into group A(hibernating myocardium group,8 dogs)and group B(sham operation group,8 dogs).In group A,thoracotomy ligation was used to cause left anterior descending artery stenosis(75%).Outdoor activities were maintained for 4 hours every day after surgery.Chronic ischemia in the target blood supply area was observed to prepare A chronic hibernating myocardial model.In group B,the silk thread passed under the anterior descending branch,but was not ligated.Both groups were terminated at 3 months after surgery.Echocardiographic dobutamine load test was used to observe left ventricular wall movement in group A,and echocardiography was used to observe systolic ventricular wall thickening rate(WT%),left ventricular ejection fraction(LVEF)and other indicators in the two groups.Results Abnormal left ventricular wall movement,WT%,LVEF were significantly reduced in group A(hibernating myocardium group).Conclusion The method of thoracotomy ligation,supplemented with regular outdoor exercise after the operation,is simple,and has a high success rate with low technical requirements for the experimenter,safe and no radiation.It is a simple and feasible method for the preparation of hibernating myocardial model.
