摘要
实验采用核磁共振(1H NMR)代谢组学方法,对MFC荷瘤小鼠和环磷酰胺(CTX)干预MFC荷瘤小鼠的肝脏提取物进行代谢谱分析,结合PCA和OPLS-DA多元数据分析方法,分析差异性代谢物和代谢通路,探索CTX抗肿瘤的作用机制。在肝脏提取物中分析确定了6个潜在的差异代谢物:丙酮酸、羟脯氨酸、肌苷、烟酰胺、胆碱、甘油磷酸胆碱。CTX的抗肿瘤作用可能与调节氨基酸代谢、能量代谢、核酸代谢和脂质代谢等代谢通路有关。
Nuclear magnetic resonance(1H NMR)was used to analyze the metabolic spectrum of liver extract from MFC tumor bearing mice and cyclophosphamide(CTX)intervention in MFC tumor bearing mice,to find out the differential metabolites and related metabolic pathways by multivariate data analysis including PCA and OPLS-DA and to explore the anti-tumor mechanism in CTX.Six differential metabolites were identified in the liver extract after CTX administration,including pyruvate,hydroxyproline,inosine,niacinamide,choline and glycerophosphorylcholine.The anti-tumor effect of CTX may be related to the regulation of amino acid metabolism,energy metabolism,nucleic acid metabolism and lipid metabolism.
