摘要
蓖麻的花序是与其产量直接相关的性状。为分析蓖麻单雌、标雌、两性系花序不同时期基因组DNA甲基化差异、寻找花序发育相关基因,本试验对4叶期、5叶期、主茎穗期及二级分枝开花期三类材料进行了甲基化敏感扩增多态性(methylation sensitive amplification polymorphism,MSAP)分析。结果表明不同发育时期3种花序的甲基化存在差异,在4叶期、5叶期、主茎穗期、二级分枝开花期的总甲基化率分别为25.26%~31.95%、14.21%~16.35%、16.58%~17.32%和21.9%~27.68%,半甲基化率分别为13.31%~15.50%、6.96%~8.93%、9.11%~9.99%和10.06%~12.58%,全甲基化率分别为11.96%~16.46%、7.08%~7.42%、6.70%~9.33%和10.69%~15.10%;此外,发现了4个可能与蓖麻花序发育相关的基因,同源序列分别编码过氧化物酶体生物合成蛋白6(LOC8287160)、GABA转运蛋白(LOC8287746)、多效耐药蛋白1(LOC8284842)和磷脂酰肌醇4-磷酸5-激酶(LOC8274135),为进一步研究蓖麻花序发育过程中DNA甲基化状况提供了参考。
The castor inflorescence is a direct trait that related to its yield. To analyze the difference of genomic DNA methylation in inflorescences of castor single female, standard female and hermaphrodite at different stages,and search for the genes that regulate the development of inflorescence. In this experiment, methylation sensitive amplified polymorphic(MSAP) analysis was applied to three materials: 4 leaves period, 5 leaves period, main stem panicle period and secondary branching period. The results showed that there were differences in methylation of three inflorescences at different developmental periods: all the methylation rates of the three inflorescence types at the 4 leaves period, 5 leaves period, main stem panicle period, and secondary branching period were 25.26%~31.95%,14.21%~16.35%, 16.58%~17.32%, 21.9%~27.68%, and the hemimethylation ratios were 13.31%~15.50%, 6.96%~8.93%, 9.11%~9.99%, 10.06%~12.58%, in addition, the total methylation rates were 11.96%~16.46%, 7.08%~7.42%, 6.70%~9.33%, 10.69%~15.10%. Four genes that may be related to the development of castor inflorescence were identified, and the homologous sequences respectively coded peroxisome biosynthesis protein 6(LOC82877-46), multidrug resistant protein 1(LOC8284842) and phosphatidylinositol 4-phosphate 5-kinase(LOC8274135). It provides a reference for further study of DNA methylation during the development of castor inflorescence.
