NSCLC癌组织及癌旁组织Rac3表达情况及其与临床预后的关系 被引量:1

EXPRESSION OF RAC3IN NSCLC CANCER TISSUESAND ADJACENT TISSUES AND ITS RELATIONSHIP WITH CLINICAL PROGNOSIS

摘要 目的研究非小细胞肺癌(NSCLC)癌组织及癌旁组织Ras相关的C3肉毒素底物(Rac)3表达情况,并分析其与预后的关系。方法标本来源于我院病理科2013年1月-12月的存档蜡块,包括106例NSCLC组织和48例癌旁正常肺组织。采用Western blot检测Rac3蛋白相对表达量,免疫组化法测定Rac3表达水平,并分析其与临床预后的关系。结果在NSCLC癌组织、癌旁组织中均检测到Rac3蛋白表达,其相对表达量[(0.87±0.19)vs.(0.55±0.14)]比较差异有统计学意义(t=10.447,P<0.05);Rac3主要表达于细胞浆或胞膜,其在NSCLC癌组织、癌旁组织中的高表达率(50.94%vs.18.75%)比较差异有统计学意义(χ^2=14.165,P<0.05)。Ⅱ~Ⅲ期患者癌组织Rac3高表达率显著高于Ⅰ期,有淋巴结转移者癌组织Rac3高表达率显著高于无淋巴结转移者,差异有统计学意义(χ^2=37.991,15.954,P<0.05)。NSCLC癌组织Rac3高表达、低表达者5年生存率(27.78%vs.67.31%)比较差异有统计学意义(log-rankχ^2=16.611,P<0.05),Rac3高表达的患者术后生存时间与低表达者比较明显缩短(P<0.05)。COX比例风险回归模型分析显示,TNM分期(RR=5.118,95%CI:3.160~9.855),淋巴结转移(RR=1.956,95%CI:1.148~3.394)及Rac3高表达(RR=2.063,95%CI:1.335~4.251)是影响NSCLC患者预后的因素(P<0.05)。结论与癌旁组织比较,NSCLC癌组织中Rac3呈高表达,且与TNM分期、淋巴结转移及预后密切相关。 Objective To investigate the expression of Ras-associated C3 botulinum substrate(Rac3) in non-small cell lung cancer(NSCLC) and adjacent tissues,to analyze its relationship with prognosis.Methods Specimens were collected from one hundred and six NSCLC tissues and forty-eight normal lung tissues in Department of Pathology of The First People′s Hospital of Kunshan from January 2013 to December 2013.Western blot was used to detect the relative expression of Rac3 protein,and immunohistochemical method was used to detect the expression of Rac3,and the relationship between Rac3 protein and clinical prognosis was analyzed.Results The expression of Rac3 protein was detected in NSCLC cancer tissues and adjacent tissues,and the relative expression((0.87 ±0.19) vs.(0.55±0.14)) had statistical significance(t=10.447,P<0.05);Rac3 was mainly expressed in cytoplasm or cell membrane,and its high expression rate in NSCLC tissues and adjacent tissues(50.94% vs. 18.75%) was significantly different(χ^2=14.165,P<0.05).The high expression rate of Rac3 in cancer tissues of stage II-III patients was significantly higher than that of stage I patients.The high expression rate of Rac3 in cancer tissues with lymph node metastasis was significantly higher than that in non-lymph node metastasis patients(χ^2=37.991,15.954,P<0.05).The 5-year survival rate of patients with high and low expression of Rac3 in NSCLC was 27.78% vs. 67.31%(log-rankχ^2=16.611,P<0.05).The survival time of patients with high expression of Rac3 was significantly shorter than those with low expression(P<0.05).COX proportional risk regression model analysis showed that TNM stage(RR=5.118,95%CI:3.160-9.855),lymph node metastasis(RR=1.956,95%CI:1.148-3.394) and high expression of Rac3(RR=2.063,95%CI:1.335-4.251) were the prognostic factors of NSCLC patients(P<0.05).Conclusion Compared with adjacent tissues,Rac3 is highly expressed in NSCLC,which is closely related to TNM stage,lymph node metastasis and prognosis.
