石榴皮鞣质对前列腺癌细胞侵袭转移影响的研究

Effect of pomegranate tannin on invasion and metastasis of prostate cancer cells

目的探讨石榴皮鞣质对人前列腺癌PC-3细胞侵袭转移的影响。方法以人前列腺癌PC-3细胞为靶细胞,通过划痕愈合实验检测石榴皮鞣质对体外培养的PC-3细胞运动能力的影响;采用Transwell小室实验(无基质胶)检测石榴皮鞣质对PC-3细胞运动能力的影响,采用Transwell小室实验(有基质胶)检测石榴皮鞣质对PC-3细胞侵袭能力的影响;采用裸鼠肺转移模型检测石榴皮鞣质对PC-3细胞体内侵袭能力的影响。结果划痕愈合实验结果显示,石榴皮鞣质能够抑制PC-3细胞的运动性,随着石榴皮鞣质浓度增加,其抑制PC-3细胞运动能力也逐渐增强,呈明显的浓度依赖性,与空白对照组比较差异有统计学意义(P<0.05);Transwell小室实验(无基质胶)显示,石榴皮鞣质能够抑制PC-3细胞运动;与空白对照组相比,随着石榴皮鞣质浓度增加,其降低PC-3细胞运动性也逐渐增强(P<0.05);Transwell小室(有基质胶)侵袭实验显示,与空白对照组相比,含有不同浓度石榴皮鞣质的实验组中,PC-3细胞穿膜细胞数即抑制其侵袭性得到有效抑制,与空白对照组相比,随着药物浓度的增加,其抑制侵袭性能力也逐渐增强(P<0.05);此外,裸鼠肺转移模型结果显示,石榴皮鞣质能够抑制PC-3细胞的侵袭性;与空白对照组相比,随着石榴皮鞣质浓度增加,PC-3细胞形成的肺转移瘤减少(P<0.05)。结论石榴皮鞣质对人前列腺癌PC-3细胞侵袭转移具有抑制作用,能抑制前列腺癌的侵袭转移。

Objective To investigate the effect of pomegranate tannin on invasion and metastasis of human prostate cancer PC-3 cells.Methods Using human prostate cancer PC-3 cells as target cells,the effect of pomegranate tannin on the migration ability of PC-3 cells in vitro was detected by scratch healing test;the effect of pomegranate tannin on the migration ability of PC-3 cells was detected by Transwell chamber test(without matrix gum);the effect of pomegranate tannin on the invasion ability of PC-3 cells was detected by Transwell chamber test(with matrix gum).The effect of pomegranate tannin on invasion ability of PC-3 cells in vivo was detected by nude mice lung metastasis model.Results Scratch healing experiment showed that pomegranate tannin could inhibit the movement of PC-3 cells.With the increase of pomegranate tannin concentration,pomegranate tannin inhibited the movement of PC-3 cells in a concentration-dependent manner,which was significantly different from that of control group(P<0.05).Transwell experiment(without matrix gel)showed that pomegranate tannin could inhibit the movement of PC-3 cells;compared with control group,pomegranate tannin inhibited the movement of PC-3 cells in a concentration-dependent manner(P<0.05).Increasing the concentration of pomegranate tannin could decrease the motility of PC-3 cells(P<0.05).Transwell(with matrix gel)invasion experiment showed that the number of PC-3 cells which penetrated the membrane was inhibited in the experimental groups with different concentrations of pomegranate tannin,and the invasion of PC-3 cells was inhibited.Compared with control group,the ability of inhibiting invasion of PC-3 cells increased with the increase of drug concentration(P<0.05).In addition,the results of lung metastasis model in nude mice showed that pomegranate tannin could inhibit the invasion of PC-3 cells.Compared with control group,the number of lung metastases formed by PC-3 cells decreased with the increase of pomegranate tannin concentration(P<0.05).Conclusion The pomegranate tannin can inhibit the invasion and metastasis of human prostate cancer PC-3 cells and the prostate cancer.
