千山活血膏治疗大鼠急性软组织损伤的作用研究

Effect of Qianshan Huoxue Plaster on acute soft tissue injury in rats

目的探究千山活血膏治疗大鼠急性软组织损伤的疗效及其作用机制。方法用自制软组织打击器建立急性软组织损伤大鼠模型,90只雄性SD大鼠随机分为模型组,扶他林(0.5 g/只/天)组和千山活血膏(0.5 g/只/天)组,每组30只大鼠,1天给药1次,连续15 d。造模后第1、4、7、15天分别记录各组大鼠的活动状态、体重,并进行损伤证候指数评分。酶联免疫法检测各组大鼠软组织损伤处炎症因子肿瘤坏死因子α(TNF-α)、白细胞介素-1β(IL-1β)、前列腺素E 2(PGE 2)的含量变化,HE染色进行组织病理学检查。结果千山活血膏可显著改善大鼠急性软组织损伤,并且软组织损伤并未对大鼠体重造成影响。由三组间的损伤评分,组织切片以及染色结果可以看出,千山活血膏与扶他林对于大鼠的软组织损伤有良好的治疗效果。同时,从炎症因子的检测结果可以看出,较模型组,千山活血膏与扶他林用药后均显著抑制了肿瘤坏死因子α、白细胞介素-1β的表达(P<0.01),且两组间效果相当。对于前列腺素E 2含量却无影响,3组间差异无统计学意义。结论千山活血膏对治疗大鼠急性软组织损伤具有较好的疗效,其机理可能是通过调控肿瘤坏死因子α、白细胞介素-1β炎症因子改善急性软组织损伤。

Objective To explore the therapeutic effect and mechanism of Qianshan Huoxue Plaster on the treatment of acute soft tissue injury in rats.Methods A rat model of acute soft tissue injury was established by a self-made soft tissue attacker.90 male SD rats were randomly divided into a model group,a Dichofenac group(0.5 g/mouse/day)and a Qianshan Huoxue Plaster group(0.5 g/mouse/day).30 rats in each group were administered once a day for 15 days.On the 1st,4th,7th,and 15th day after modeling,the activity status and body weight of each group were recorded,and the injury syndrome index was scored.The levels of inflammatory factors TNF-α,IL-1βand PGE 2 in soft tissue injury of rats in each group were detected by enzyme-linked immunosorbent assay(ELISA).Histopathological examination was performed by HE staining.Results Qianshan Huoxue Plaster can significantly improve acute soft tissue injury in rats,and soft tissue injury does not affect the body weight of rats.From the injury scores,tissue sections and staining results of the three groups,it can be seen that Qianshan Huoxue Plaster and Dichofenac have a good therapeutic effect on soft tissue injury in rats.At the same time,the results of inflammatory factors suggested that Qianshan Huoxue Plaster and Dichofenac significantly inhibited the expression of TNF-αand IL-1β(P<0.01)compared with the model group,and there was no significant difference between the two groups.However,there were no effects on PGE 2 content and no significant difference between the 3 groups.Conclusion Qianshan Huoxue Plaster had good effect on the treatment of acute soft tissue injury in rats,which may ameliorate acute soft tissue injury by regulating the expression of TNF-αand IL-1β.
