摘要
目的探讨黄芪多糖(APS)对双环己酮草酰二腙(CPZ)诱导的急性脱髓鞘小鼠模型的保护作用。方法复制CPZ小鼠模型,将54只C57BL/6雌性小鼠随机分为三组:正常组、CPZ组和APS组,每组18只。正常组小鼠予以普通饲料,CPZ组和APS组喂食含0.2%CPZ的混合饲料建模,连续6 w。从第2周起,APS组按500 mg/(kg·d)腹腔注射APS,CPZ组给予等量生理盐水,持续给药至实验结束。记录小鼠的体重和进食量,分别取实验4、5和6 w小鼠的脑组织称重并进行髓鞘固蓝染色、苏木素-伊红(HE)染色和免疫荧光组织化学双染,观察APS对CPZ小鼠脑组织形态学、髓鞘脱失和髓鞘碱性蛋白(MBP)表达的影响。结果APS能增加CPZ小鼠体重、进食量和小鼠全脑重量,降低小鼠脏器系数;APS能有效改善CPZ小鼠脑组织的髓鞘脱失,增加MBP的表达;形态学观察可见CPZ组小鼠脑组织小胶质细胞聚集,数量增多,APS抑制小胶质细胞在脑内的聚集。结论APS对CPZ诱导的急性脱髓鞘小鼠具有神经保护作用。
Objective To investigate the protection of Astragalus Polysaccharide(APS)on acutely demyelinated mice model induced by cuprizone(CPZ).Methods Fifty-four female C57BL/6 mice were randomly divided into three groups:normal,CPZ model and APS-treated groups with 18 mice in each group.Mice in normal group were fed with normal diet for 6 weeks,mice in CPZ group and APS group were fed with diet containing 0.2%CPZ for 6 weeks.Mice in APS group were given intraperitoneal injection of APS 500 mg/(kg·d)and CPZ model mice were injected with the same amount of saline from the second week until the end of the experiment.To observe the effect of APS,the body weight and food intake of mice were recorded.Brains of mice of 4~6 weeks were obtained for Luxol fast blue(LFB)staining and hematoxylin/eosin(HE)to observe the morphological changes and demyelination.Immunofluorescence histochemical staining was used to detect the expression of myelin basic protein(MBP)in brain of mice.Results APS could increase body weight,food intake and whole brain weight of CPZ mice,reduce organ coefficients of mice.APS effectively improved the demyelination of brain tissue and increased the expression of MBP protein in CPZ mice.Morphological observation showed that microglia aggregated and increased in the brain of CPZ model mice.APS inhibited microglia aggregation in the brain.Conclusions APS has neuroprotective effect on acutely demyelinating mice induced by CPZ.
